摘  要:目的:研究不同干燥方法对北青龙衣萘醌类成分含量的影响,为建立适合北青龙衣产地加工方法提供参考。方法:采用晒干,阴干,40、50、60℃烘干,冷冻干燥,微波干燥5种方法对北青龙衣样品进行干燥处理。采用UPLC-Triple TOF-MS/MS技术对样品进行测定,数据用Marker View软件进行主成分分析,化合物通过Peak View2.0/masterview1.0软件,依据精确质量数和同位素峰度比确定分子式,再通过对照品及数据库的二级谱图比对、裂解规律分析,结合已有文献报道,确定结构式。结果:不同干燥方法萘醌类有效成分含量发生了明显的变化,鉴定或推断了北青龙衣中24个萘醌类化合物结构,确定了40℃烘干为最适宜干燥方法。结论:该方法快速、准确,可为北青龙衣不同干燥方法的化学成分鉴定与分析提供一种新的策略,对北青龙衣加工生产具有一定的指导意义。Objective:To study the influence of different drying methods on the naphthoquinone components in Juglans mandshurica.Methods:Fresh Juglans mandshurica sample was processed by five drying methods including drying in the sun,drying in the shade,forced air drying with different temperature,freeze drying and microwave drying.Samples of Juglans mandshurica from five drying methods were assessed by UPLC-Triple TOF-MS/MS.The Marker View software was used for data processing and multivariate statistical analysis.The formula of compounds were determined by accurate mass and isotopic abundance ratio from target screening function of Peak View 2.0/masterview1.0 software.Then their structure were determined by analysis of MS/MS fragment or comparison with standard substances and references.Results:Naphthoquinones were the major markers in samples of Juglans mandshurica from different drying methods.24 naphthalenequinones were identified or inferred in Juglans mandshurica.Forced air drying with 40 ℃ was considered as the most suitable drying method.Conclusion:UPLC-Q-TOF/MS method develops a new strategy can identify and analyze chemical constituents from different drying methods rapidly and accurately,which will be serviced in the large scale processing of Juglans mandshurica.
