摘  要:目的:研究白及小分子药效组分对二氧化硅诱导的实验性矽肺大鼠肺纤维化抑制活性及其可能分子机理。方法:95%乙醇回流提取,聚酰胺柱色谱法富集制备白及小分子药效组分,SiO_2气管滴注法建立大鼠矽肺模型。通过大鼠肺指数变化、HE切片观察及HYP含量分析药效活性;通过测定血清SOD、MDA、NF-κB、IL-1β、PDGF、TGF-β_1、TNF-α含量变化及流式细胞术测定CD4^+/CD8^+T细胞比值,分析白及小分子药效组分可能的作用机理。结果:与模型组比较,白及小分子药效组分低(20 mg/kg)、高(40 mg/kg)剂量均可显著升高血清SOD活性,降低MDA及NO含量,并逆转CD4^+/CD8^+比值(P<0.01),同时还可剂量依赖性降低血清各炎性细胞因子含量,从而有效降低血清HYP及肺指数,改善肺组织结构,抑制纤维化进程。结论:白及小分子药效组分对矽肺大鼠机体抗氧化系统、免疫系统及炎性细胞因子水平均具有良好的调节作用,从而有效抑制肺纤维化,是中药白及治疗矽肺的药效活性组分。Objective:To approach the anti-pulmonary fibrosis effect and the possible molecular mechanism of the small molecule components of Bletilla striata(SMCBS). Methods:SMCBS was prepared by 95% ethanol reflux extraction method and further enriched by polyamide column chromatography. Rat silicosis model was established by invasive intratracheal instillation method. The effect and molecular mechanism of the SMCBS was evaluated by lung index, lung pathological section, the content of HYP, and the level of SOD, MDA,NF-κB,IL-1β,PDGF,TGF-β1 ,TNF-α were detected,and the CD4^+ ,CD8^+ T lymph cell and CD4^+/CD8^+ ratio in serum were detected by flow cytometry. Results:Compared with the model group, both low (20 mg/kg)and high (40 mg/kg)dosage SMCBS treat- ment groups could remarkably elevate the serum SOD level and lower the MDA, NO level, and effectively reverse the CD4 ^+/CD8 ^+ ratio (P 〈 0. 01 );moreover, SMCBS can dose dependently decrease all the inflammatory cytokines, and lower HYP content in serum. HE pathological section confirmed that both low and high dosage SMCBS treatment group can effectively prevent lung fibrosis and lower the lung index. Conclusion:The small molecule components of Bletilla striata can effectively prevent lung fibrosis though regulating the anti- oxidation system,immune system and cytokine level;SMCBS is one of the active components of Bletilla striata on silicosis therapy.
