摘 要:为了防止楸子(Malus prunifolia Willd.Borkh)初代组织培养外植体褐化,设置选用不同外植体、消毒方法、培养温度、光照、6-BA浓度及不同防褐化剂类型与浓度研究楸子离体培养过程中外植体褐化影响因素。结果表明,选用当年生新枝上1~3cm的带芽茎段作为材料,使用0.1%HgCl_2消毒,4℃低温暗培养5d,并在培养基中添加1~2g/L抗坏血酸或0.5~2g/L聚乙烯吡咯烷酮及0.5mg/L的6-BA可以有效防止褐化,且植株生长旺盛,色泽正常,生长势较好。In order to prevent explant from browning in early generation culture of Malus prunifolia Willd. Borkh, the effects of different kinds of explants , disinfection methods, culture temperature,illumination, concentrations of 6-BA and browning agent on explant browning tissue culture were selected and studied. Results show that 1~3 cm as explant ma- terial containing buds of stem section can be prevent from browning better, by using 0. 1 ~ HgCl2 to disinfect, culturing under 4 ℃ for 5 days. The concentration of 1~ 2 g/L of Vc or 0. 5 ~ 2 g/L of PVP supplemented to medium can be in- hibited explant from browning; The concentration of 0.5 g/L of browning and result in a good growth 6- B.A. supplemented to medium can reduce exptant
