摘 要:目的:探讨中药姜黄素对肝癌HepG2细胞增殖及凋亡的影响。方法:将肝癌HepG2细胞分为姜黄素组与空白对照组,采用四甲基噻唑氮蓝(MTT)比色法检测不同浓度(10μmol/L、20μmol/L、40μmol/L、60μmol/L和80μmol/L)姜黄素处理后HepG2细胞的增殖抑制率,流式细胞术检测姜黄素对HepG2细胞周期及细胞调亡的影响。结果:不同浓度姜黄素组中,随着药物浓度增加,作用时间延长,细胞的增殖抑制率升高,组间比较差异具有统计学意义(P<0.05)。与空白对照组比较,80μmol/L姜黄素组S期细胞比例显著增加,细胞凋亡率明显升高,差异均有统计学意义(均P<0.05)。结论:姜黄素可以有效抑制肝癌HepG2细胞的增殖,并促进细胞凋亡,其抑癌作用可能与阻滞细胞于S期有关。Objective:To explore the effect of traditional Chinese medicine curcumin on HepG2 live cancer cells proliferation. Methods: The HepG2 cells were divided into acurcumin group and a control group. The inhibitory rate of proliferation of HepG2 cells treated with different concentration of curcumin (10, 20, 40, 60 and 80 μmol/L) were analyzed by MTT assay. The influence of curcumin on HepG2 live cancer cells cycle and cells apoptosis were analyzed by flow cytometry. Results: In the different concentrations of curcurain group, the inhibitory rate was increased with the increase of drug concentration and the exposure time, and the difference among groups were statistically significant ( P 〈 0. 05). Compared with the control group, the proportion of S phase cells in 80 μmol/L curcumin group was significantly increased, the apoptosis rate was significantly increased ( P〈0.05). Conclusion: Curcumin could inhibit the proliferation of hepatocellular carcinoma HepG2 cells and accelerate the apoptosis via the inhibition of cell cycle.
