热休克蛋白60在高脂及高脂高糖大鼠睾丸中的表达及对生精细胞凋亡的影响

摘　　要：目的探讨热休克蛋白60(Hsp60)在高脂及高脂高糖SD大鼠睾丸组织中的表达情况及对生精细胞凋亡的影响。方法将雄性SD大鼠60只随机分成对照组15例、高脂组15例及高脂高糖组30例,对照组给予普通饲料喂养,高脂组给予高脂饲料喂养,24周后加入含0.2%丙硫氧嘧啶继续喂养;高脂高糖组给予高脂饲料喂养,24周后腹腔注射链脲佐菌素,所有大鼠均喂养36周。造模成功后取睾丸组织,用免疫组化法和免疫印迹试验分析检测大鼠睾丸组织中Hsp60表达情况,并检测组织中细胞凋亡情况。结果对照组生精细胞数量多且按序逐层排列,管腔中央可见大量精子细胞,HSP60少量表达于次级精母细胞;高脂组及高脂高糖组生精细胞数量减少且排列紊乱,部分切片可观察到生精细胞脱落。免疫组化及免疫印迹试验结果均显示,高脂组及高脂高糖组Hsp60蛋白表达量高于对照组(P<0.05),而高脂组与高脂高糖组间比较差异无统计学意义(P>0.05)。高脂组及高脂高糖组的生精细胞凋亡数量明显多于对照组,且高脂高糖组生精细胞凋亡数量多于高脂组(P<0.05)。结论高脂及高脂高糖大鼠睾丸生精细胞凋亡的发生可能与Hsp60密切相关。Objective To investigate the expression of heat shock protein 60（Hsp60） in the testis of SD rats with hyperlipemia or hyperlipemia combined with hyperglycemia, and its effect on the apoptosis of spermatogenic cells. Methods Sixty male SD rats were ramlomly divided into control group （ n = 15 ） , hyperlipemia group （ n = 15 ） , and hyperlipemia combined with hyperglycemia group （n= 30）. The control group received a normal diet. The hyperglycemia group was given 0.2% propyhhiouraeil additionally after 24 weeks of high-fat diet. Iutraperitoneal injection of streptozotoein was conducted in the hyperlipemia combined with hyperglycemia group after 24 weeks of high-fat diet. All rats were fed for 36 weeks. The testis tissue was obtained after successtul modeling. The expression of Hsp60 protein in the testis of the rats was detected by immunohistoehemistry and Western blot, and the apoptosis of spermatogenie cells was assessed in the testicle tissues. Results In the eontrol group, a large number of spermatogenic cells were observed and arranged in sequence, a lot of sperms were fnund in the center of the lumen, a few HSP60 expressed in the seeondatw spermatoeyte. In the hyperlipemia group and the hyperlipemia combined with hyperglycemia group, the number of spermatogenie cells decreased and the arrangement of spermatogenic eells was disordered, spermatogenic cells shedding was observed in some slides. The results of immunohistnehemislry and Western hlot showed that the expression level of Hsp60 protein in the hyperlipemia group or the hyperlipemia combined with hyperglycemia group was higher than that in the control group（ P 〈 0.05 ） , but there was no significant difference between the hyperlipemia group and the hyperlipemia combined with hyperglycemia group（P 〉0.05）.
