摘 要:目的:观察柴芪汤对饮食诱发的代谢综合征(MS)模型大鼠血管损伤的预防及治疗的干预效果,探讨其血管保护机制。方法:将56只SPF级雄性SD大鼠随机分为正常组、模型组、中药预防组和中药治疗组,除正常组外其余各组用高盐高脂高糖饮食喂养。中药预防组从造模第一天开始给予柴芪汤[5.67 g/(kg·d)];中药治疗组前8周使用蒸馏水灌胃对照,从第8周开始使用柴芪汤灌胃8周。分别于8周末和16周末检测大鼠血清血糖及胰岛素,评价胰岛素抵抗情况,同时采用ELISA法检测各组大鼠血清中s VCAM-1、s ICAM-1、TNF-α及IL-6的水平,并取大鼠腹主动脉血管检测NF-κB蛋白表达。结果:模型组大鼠血糖、胰岛素、胰岛素抵抗指数、血脂、血清s VCAM-1、s ICAM-1、TNF-α及IL-6水平、血管NF-κB蛋白表达明显高于正常组(P<0.05);使用柴芪汤干预后各项指标均明显低于模型组(P<0.05);但早期预防组的各项指标较治疗组的更具有优势。结论:柴芪汤可能通过影响血清s VCAM-1、s ICAM-1、TNF-α、IL-6的水平及NF-κB蛋白从而对MS血管损伤起到一定的干预效果,同时柴芪汤早期预防用药比治疗给药效果明显。Objective :To observe the effect of Chaiqi decoction on vascular injury of metabolic syndrome rat models and to investigate its mechanism. Methods:The rats were randomly divided into a normal group, a model group, a prevention group and a treatment group. The rat models were established by feeding high fat, high sugar and high salt diet for 8 weeks. The prevention group was given Chaiqi decoction [ 5.67 g/( kg ·d) ] from the beginning of modeling, and the treatment group was given Chaiqi decoction since the 8th week. The blood glucose and insulin were detected at 8th and 16^th weeks. The levels of sVCAM- 1 ,sICAM- 1 ,TNF- α and IL - 6 in serum were measured by ELISA, and the protein expressions of NF - κB in the vessel were detec ted by Western blot. Results: Compared with that of the normal group, the blood glucose, insulin, insulin resistance index, the levels of sVCAM - 1, sICAM - 1, TNF -α and IL - 6 in serum, the protein expressions of NF - κB in vessel were significantly increased in model group ( P 〈0. 05 ). All the indexes of the prevention group and treatment group were lower than those of the model group, and the difference was statistically significant (P 〈 0.05 ). The indexes of prevention group had more advantages than those of the treatment group. Conclusion: Chaiqi decoction has some active effects on the MS vessel injury by reducing the levels of sVCAM - 1, sICAM - 1, TNF - α and IL - 6 in serum and the protein expressions of NF - κ in vessel. Using Chaiqi decoction at early stage as a prevention can have a better result than as a treatment.
