摘 要:目的:研究AAV9-SERCA 2a基因转染对急性心房颤动(房颤)动物模型心房肌细胞上L型钙通道表达的影响。方法:采用60只新西兰大白兔为实验对象,采用心包注射法转染AAV9-SERCA 2a基因;将实验对象随机分为4组:房颤组、转荧光蛋白房颤组、转SERCA 2a基因房颤组和对照组。转染后建立急性房颤模型,采用膜片钳及荧光免疫组织化学法检测心房肌细胞上L型钙通道的电流变化及其L型钙通道蛋白表达的情况。结果:转SERCA 2a基因房颤组心房肌细胞上LVDCCa1c蛋白表达明显高于房颤组(P<0.05),与对照组比较差异无统计学意义。转SERCA 2a基因房颤组心房肌细胞上L型钙通道电流密度高于房颤组,差异有统计学意义(P<0.05),与对照组比较差异无统计学意义。结论:转SERCA 2a基因治疗可减少急性房颤模型心房肌细胞钙超载,减少房颤对L型钙通道电流密度的影响,对防止心脏电重构具有积极的意义。Objective:To study the effects of AAV9 - SERCA 2a gene transfection on L type calcium channel expression of atrial myocytes in animal models with acute atrial fibrillation. Method: Sixty New Zealand white rab- bit were selected as experimental objects, the pericardium injection was used to transfect AAV9 - SERCA 2a gene. The subjects were randomly divided into 4 groups: atrial fibrillation group, AAVg-EGYP group, SERCA 2a group and control group. Acute atrial fibrillation models were established after transfection. Patch clamp and fluorescent immuaohistochemical method were used to detect L type calcium channel current changes and expression. Result: LVDCCalc protein expression on atrial myocytes was obviously higher in SERCA 2a group than that in atrial fi- brillation group (P〈0.05), and no statistically significant difference was observed between SERCA 2a group and control group. The L type calcium channel current density was higher in SERCA 2a group than that in atrial fibril- lation group (P〈O. 05), and no statistically significant difference was observed between SERCA 2a group and control group. Conclusion: The therapy of SERCA 2a gene transfection can reduce the calcium overload on atrial myocytes in acute atrial fibrillation models, and reduce the effect of atrial fibrillation on L type calcium channel current density, which has a positive influence to prevent cardiac electrical remodeling.
