马骨关节炎模型关节液中MMP-2、MMP-9、MMP-13含量变化的研究

摘　　要：为了探究患骨关节炎的马关节液中生物指标——基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶-13(MMP-13)的含量变化关系,试验选用13匹蒙古马随机分为试验组(n=8)和对照组(n=5),试验组左侧腕关节内注入2 m L两性霉素B(25 mg/m L)建立马骨关节炎模型,对照组左侧腕关节注射2 m L生理盐水,每周采集关节液1次,用ELISA法检测马关节液中MMP-2、MMP-9、MMP-13的含量。结果表明:试验组第1周起开始出现不同程度跛行;与对照组相比,试验组关节液MMP-2、MMP-9、MMP-13的含量分别从第3,1,2周开始显著增加(P<0.05),且均保持增高趋势直至第9周试验结束。说明随骨关节炎加重MMP-2、MMP-9和MMP-13含量呈上升趋势,对基质的分解作用增强,造成软骨基质破坏。其中MMP-9初期增加比较显著,可以作为临床快速检测马骨关节炎的重要指标。To investigatethe changes of synovial fluid matrix metalloprotcinase - 2 （ MMP - 2 ）, matrix metalloproteinase - 9 （ MMP - 9 ）, matrix metallopreteinase - 13 （ MMP - 13 ） in osteoarthritis - affected equine, 13 Mongolian horses were selected and randomly divided into experimen- tal group （ n = 8） and control group （ n = 5 ）. To establish a horse asteoarthritis model,the left carpal joint in the experimental group was injected by 2 mL amphotericin B （25 mg/mL） ;the control group was injected into the left carpal joint with 2 mL saline. Synovial fluid was collected once a week, and the concentrations of MMP-2, MMP-9 and MMP -13 were measured by using ELISA kit. The results showed thatcom- pared with the control group, the synovial fluid of MMP-2,MMP-9, and MMP- 13 in experimental group significantly increased （P 〈 0.05 ）, respectively, from the 3th, lst,2nd week, and keep the increasing trend until the ninth weed. The results demonstrated that the in- crease of thecontent of MMP -2,MMP - 9 and MMP - 13 enhanced decomposition of the matrix, resultingin damage to the cartilage matrix. Wherein, the content of MMP - 9 increased more significantly at early course, which is a potential biomarker as an important indicator forrapid diagnose of horse osteoarthritis.
