FASN基因的多态性及其与中国荷斯坦奶牛产奶性状的相关性分析

摘　　要：为了分析脂肪酸合成酶(fatty acid synthase,FASN)基因的多态性及其与中国荷斯坦奶牛产奶性状的相关性,试验选取230头荷斯坦奶牛,采用限制性片段长度多态性聚合酶链反应(Restriction fragment length polymorphism-polymerase chain reaction,RFLP-PCR),对FASN基因3'-UTR区和外显子部分片段的多态性进行检测,并对突变位点与奶牛产奶性状的相关性进行分析。结果表明:在所分析的片段中,检测到3'-UTR区存在一个C→T碱基的置换突变(g.51403203C>T);该突变位点为Bam HⅠ酶的自然酶切位点,酶切后存在CC、TC、TT三种基因型,TT基因型的乳脂率和脂蛋白比显著高于TC基因型、CC基因型(P<0.05),而三种基因型间乳蛋白率和305 d产奶量差异不显著(P>0.05)。To investigate polymorphism of the fatty acid synthase （FASN） gene and its association with the milk production traits in Chinese Holstein dairy cows, 230 Holstein cows were choose for detection of the polymorphisms of FASN gene in 3＇ - UTR and part of exon regions by using the method of restriction fragment length polymorphism - polymerase chain reaction （ RFLP - PCR）. Moreover, the correlation of muta- tion sites with cow milk production traits was analyzed. The results showed that one C 〉 T SNPs site was detected in the 3＇ - UTR region, and the mutation site had a natural enzyme BamH I enzyme cutting sites; there are three genotypes （CC, TC, and TT） after enzyme digestion; TI＂ genotype was significantly correlated with the milk fat percentage and lipid protein ratio （P 〈 0.05 ）. However, the milk protein rate and milk yield of 305 days had no significant association （ P 〉 0.05 ）.
