摘  要:目标:分析一个被命名为miR-340的miRNA在HCC中的表达,并筛选出与FGFR4表达相关的miRNA。方法:利用miRNA生物信息学预测数据库分析,寻找肝癌转移相关的miRNA,利用RT-PCR检测miR-340在病人的肿瘤组织和癌旁肝组织中以及肝癌细胞系中的表达,利用western blot检测FGFR4表达水平。结果:miRNA生物信息学预测数据库预测结合RT-PCR和前期Western blot的结果,选择miR-340作为进一步研究的对象,通过RT-PCR和western blot实验,证明miR-340过表达显著抑制FGFR4的表达,而miR-340低表达可使FGFR4表达升高。结论:miR-340和FGFR4在肝癌中存在较为显著的负相关性。Objective:To analyze the expression of a miRNA named miR-340 in HCC and to screen miR- NAs related to FGFR4 expression.Methods.MiRNA bioinformatics prediction database analysis was used to find miRNAs related to metastasis of liver cancer.The expression of miR-340 in tumor tissues,adjacent liver tissues and hepatocellular carcinoma cells was detected by RT-PCR.The expression of FGFR4 was detected by western blot.Results:MiR-340 was selected as a further study object by miRNA bioinformatics predictive database prediction combined with RT-PCR and early western blot results.RT-PCR and western blot experi2 ment showed that miR-340 overexpression could significantly inhibit the expression of FGFR4,while the low expression of miR-340 could increase the expression of FGFR4.Conelusion: MiR-340 and FGFR4 have a significant negative correlation in liver cancer.
