六味地黄丸对阿霉素肾病小鼠足细胞nephrin和podocin表达的影响

摘　　要：目的:观察六味地黄丸干预阿霉素小鼠肾病对足细胞nephrin和podocin表达的影响,探讨其防治阿霉素肾病的作用机制。方法:27只Balb/c小鼠,分为正常组(Normal)7只、模型组(Model)10只、六味地黄丸组(LWDHP)10只,Model组和LWDHP组一次性尾静脉注射阿霉素10.5mg·kg^(-1)造模,LWDHP组给予LWDHP,Normal组和Model组给予等量生理盐水,持续灌胃28d后处死取材。生化检测小鼠24h尿总蛋白(UTP)含量以及血总蛋白(TP)、白蛋白(ALB)、尿素氮(BUN)、血肌酐(SCr)、胆固醇(CHOL)、甘油三酯(TG);光镜电镜观测肾组织病理变化;Western blotting观察肾组织nephrin及podocin的蛋白表达水平。结果:LWDHP组24h尿蛋白UTP、血TP、ALB升高;BUN、CHOL、TG均有显著降低(P<0.05);光镜和电镜显示肾脏病理改变改善;podocin和nephrin蛋白表达水平显著升高(P<0.05)。结论:六味地黄丸防治阿霉素小鼠肾病,可能与保持足细胞相关蛋白podocin和Nephrin的表达相关。Objective： To observe the effect oF Liuweidihuang Pill on the expression ot nephrm anct poaocin in adriamycin mice, and to explore its mechanism of prevention and treatment of adriamycin nephropathy. Methods： 27 Balb/c mice were divided into normal group （normal）, model group （model） 10, Liuweidihuang pill group （LWDHP） 10, Model group and LWDHP group one-time tail vein injection The rats in LWDHP group were given LWDHP, normal group and Model group were given equal volume of normal saline, and the rats were sacrificed after 28 days of continuous administration. （UB）, serum total protein （TP）, albumin （ALB）, urea nitrogen （BUN）, serum creatinine （SCr）, cholesterol （CHOL）, triglyceride （TG） The pathological changes of nephrin and podocin in renal tissue were observed by western blotting. Results： BUN, CHOL and TG were significantly decreased （P〈0.05）. The expression of podocin and nephrin protein was significantly increased by light microscopy and electron microscopy. The expression of podocin and nephrin protein in LWDHP group was significantly higher than that in control group （P〈0.05）.Conclusion： Liuweidihuang pill can prevent nephropathy in adriamycin mice, which may be related to the expression of podocin and Nephrin.
