摘  要:目的:动态观察艾灸肺俞、膏肓俞治疗肺纤维化大鼠E-钙黏蛋白(E-cad)、α-平滑肌蛋白(α-SMA)、波形蛋白(vimentin)几种上皮-间质转分化(EMT)标志物的表达。方法:选用SD雄性大鼠44只,随机分为4组:正常对照组、模型组、艾灸治疗组、泼尼松治疗组。除正常对照组外,其余三组均采用博来霉素灌入气管复制肺纤维化模型,治疗组从造模后第1天开始分别艾灸及泼尼松灌胃治疗,在造模后第11天、第23天、第35天取材。采用HE染色、masson染色观察肺组织病理变化和采用免疫组化法检测E-cad、α-SMA、vimentin的表达。结果:艾灸组、泼尼松组与同时段模型组相比,肺组织病理程度明显降低,上皮细胞标记物E-cad表达升高,间质细胞标记物α-SMA、vimentin表达降低。结论:艾灸治疗能升高E-cad水平,降低α-SMA、vimentin水平,降低肺泡炎程度及延缓肺纤维化进程。Objective : To observe the regulation effects of several epithelial - mesenchymal transition (EMT) differentiation markers such as E - cadherin adhesion protein ( E - cad ) and alpha smooth muscle protein ( α - SMA ) , vimentin ( VIM ) through moxibustion at Feishu and Gaohuangshu against the pulmonary fibrosis process of model rats. Methods : Forty - four SD male rats were recruited and divided into four groups : normal control group, model group, moxibustion group and prednisone group, Pulmonary fibrosis model was made by the injection of bleomycin into the trachea,5 mg/kg, bw. Since the second day after modeling, rats of the moxibustion group were moxibusted at Feishu and Gaohuangshu, once a day and lasted for 35 days. Rats in the prednisone group were given stomach - injection of prednisone for every day. At 11 d ,23 d and 35 d post - modeling, the rats were randomly selected and sacrificed. HE staining and Masson staining were used to observe the pathological changes of lung tissue and the ex- pression of E - cad, alphcα - SMA and vimentin were detected by immunohistochemistry. Results : Compared with the model group, the moxibustion group and the prednisonc group significantly reduced the degree of lung tissue pathology, increased the expression of epithelial marker E - cadherin, qualitative cell marker compounds α - SMA and vimentin expression decreased. Conclusion : Moxibustion treatment can increase the level of E - cad, reduce the level of - SMA, vimentin and reduce the degree of pulmonary inflammation and delay the progression of pulmonary fibrosis.
