摘  要:目的:通过观察滋阴明目丸对视网膜光损伤大鼠模型Bcl-2、p53表达的影响,探讨滋阴明目丸对视网膜光损伤大鼠模型的保护作用及治疗机制。方法:将60只Sprague-Dawley大鼠随机分为空白对照组、模型对照组、阳性对照组、滋阴明目丸低剂量组、滋阴明目丸中剂量组、滋阴明目丸高剂量组6组,每组10只。空白对照组不予处理,其余均建立视网膜光损伤大鼠模型,造模后第3天予以灌胃,连续15 d,其中滋阴明目丸低、中、高剂量组予以不同剂量滋阴明目丸灌胃,阳性对照组予以杞菊地黄丸灌胃。取大鼠眼球做免疫组化检测Bcl-2及p53的表达。结果:Bcl-2免疫组化结果:与空白对照组比较,模型对照组有统计学差异(P=0.025<0.05);与模型对照组比较,阳性对照组(P=0.003<0.01)、中剂量组(P=0.004<0.01)及高剂量组(P=0.001<0.01)均有显著统计学差异,低剂量组无明显差异(P>0.05)。p53免疫组化结果:与空白对照组比较,模型对照组有显著统计学意义(P=0.000<0.01);与模型对照组比较,低剂量组无明显差异(P>0.05);阳性对照组(P=0.012)、中剂量组(P=0.002)及高剂量组(P=0.002)均有统计学差异(P<0.05)。结论:视网膜光损伤模型可使大鼠视网膜组织抗凋亡蛋白Bcl-2表达降低,促凋亡蛋白p53表达增高,滋阴明目丸能有效增强Bcl-2的表达,降低p53的表达,从而对细胞凋亡起到双向调节作用,对视网膜光损伤模型具有一定的治疗作用。Objective:By observing Yin- Enriching and Eye -Brightening Pill on retinal light damage rats' Bcl -2 and p53 expressions to discuss the protective effect and mechanism. Methods : Sixty Sprague Dawley rats were randomly divided into blank control group, model control group, positive control group, Yin - Enriching and Eye - Brightening Pill low, middle and high dose groups,with 10 rats in each group. Do not deal with blank control group. The rest were to establish retinal light injury rat model. 3 days after modeling,the rats were given the medicine by gavage for 15 days. Yin -Enriching and Eye -Brightening Pill groups were given the corresponding doses of Yin - Enriching and Eye - Brightening Pill orally and the positive control group was given Qiju Dihuang Pills by gavage. The expressions of Bcl - 2 and p53 were detected by immunohistochemistry in rats. Results : Bcl - 2 immunohistochemistry results : Compared with the blank control group, model control group had statistical difference ( P = 0. 025 〈 0.05 ). Compared with the model control group ,the difference in high dose group and positive control group (P = 0. 003 〈 0.01 ) and middle dose group P =0. 004 〈0.01 P =0.001 〈0.01 was significant. The low dose group had no significant difference ( P 〉 0. 05 ). P53 expression : Compared with the blank control group, model control group was statistically significant P = O. 000 〈 0. 01. Compared with the model control group,low dose group had no significant difference (P 〉 O. 05 ). The positive control group ( P = 0. 012 ) , middle dose group ( P = O. 002 ) and high dose group ( P = 0. 002 ) had statistically significant differences ( P 〈 0.05 ). Conclusion : Retinal light damage model can decrease expression of anti apoptotic protein Bel - 2 and increase pro apoptotie protein p53 expression. Yin - Enriching and Eye - Brightening Pill can effectively enhance bel - 2 expression and decrease of p53 expression so as to make cell apoptosis by two - way regulat
