摘 要:目的:探讨鲜鱼腥草挥发油对哮喘豚鼠肺组织干细胞因子(SCF)和KL-6表达的影响,分析鲜鱼腥草挥发油在减轻气道炎症可能的靶点及作用机制,为鲜鱼腥草挥发油制剂的开发提供实验依据。方法:对照组豚鼠给予生理盐水灌胃。将哮喘模型豚鼠分为模型组、高剂量组、中剂量组、低剂量组四组,模型组给予生理盐水灌胃,另外三组给予不同剂量鲜鱼腥草挥发油灌胃。采用酶联免疫吸附试验(ELISA)观察鲜鱼腥草挥发油对致敏攻击豚鼠肺组织中SCF、KL-6含量的影响。结果:哮喘模型组肺组织SCF含量升高(P<0.05),与高剂量组、中剂量组、低剂量组肺组比较差异有统计学意义(P<0.05)。哮喘模型组豚鼠肺组织KL-6含量增加(P<0.05),经鲜鱼腥草挥发油干预以后,高剂量组肺组织中KL-6的含量明显下降(P<0.01),中剂量组、低剂量组与模型组比较差异无统计学意义(P>0.05)。结论:哮喘豚鼠肺组织内SCF和KL-6表达增加,与哮喘发病密切相关。鲜鱼腥草挥发油能作用于哮喘豚鼠肺组织,能够降低哮喘豚鼠肺组织SCF和KL-6的表达,具有一定的减轻哮喘气道炎性反应的作用。Objective: To investigate the effect of volatile oil from fresh herba houttuyniae on the expression of stem cell factor(SCF) in the lung tissue and KL-6 of asthmatic guinea pigs, and to evaluate the potential of volatile oil from herba houttuyniae in relieving the pathogenesis of airway inflammation and provide the experimental basis for the development of herba houttuyniae preparation. Methods: The control group was given normal saline by intragastric administration. Asthmatic guinea pigs were divided into four groups: the model group was given normal saline by intragastric administration, and the other three groups were given different doses of volatile oil from herba houttuyniae. The effects of volatile oil from herba houttuyniae on the contents of SCF and KL-6 in lung tissue of sensitized guinea pigs were observed by enzyme-linked immunosorbent assay (ELISA). Results: The content of SCF in lung tissue of model group was increased (P〈O. 05 ), there were significant differences between model group and high dose group, middle dose group, low dose group ( P〈O. 05 ). The content of KL-6 in lung tissue of model group was increased ( P〈O. 05 ), the content of KL-6 in lung tissue of high dose group was decreased (P〈0. 01 ). There were not significant differences between model group and middle dose group, low dose group (P〉0. 05 ). Conclusion : The expressions of SCF and KL-6 in the lung tissue of asthmatic guinea pigs are increased, which are closely related to the pathogenesis of asthma. The volatile oil from fresh herba houttuyniae can effectively reduce the expression of SCF and KL-6 in the lungs of asthmatic guinea pigs, and has a good clinical effect in relieving the airway inflammatory response in asthmatic guinea pigs.
