摘 要:本研究旨在通过食蟹猴经腹腔注射低剂量百草枯来探讨胰腺α-突触核蛋白(α-synuclein,α-syn)的病理变化。由广西南宁灵康赛诺科生物科技有限公司提供的12只(8~10岁)健康雄性食蟹猴为实验动物,随机分为实验组A、B和正常对照组C,每组4只,实验组分别于给药百草枯(paraquat,PQ)5个月,2个月后安乐死(对照组每阶段2只)。利用免疫组织化学方法和免疫蛋白印记方法检测各组食蟹猴胰岛α-syn的表达特点和表达量。结果显示,α-syn免疫活性物质主要见于胰岛细胞胞浆,并且α-syn在各组食蟹猴胰岛存在差异表达,其中给药百草枯5个月后的食蟹猴胰岛α-syn表达量最高,对照组表达量最低。我们的研究表明食蟹猴腹腔注射低剂量百草枯可致胰岛α-syn表达上调,百草枯是造成胰岛腺α-syn差异表达的关键因素之一,其结论将为相关疾病的模型制作、早期诊断及干预提供新的思路。This study aimed to explore the pathological changes of α-synuclein (α-syn) in pancreas by injecting small dose ofparaquat into cynomologus monkeys. 12 healthy male cynomolgus monkeys (8-10 years old), provided by the Wincon Thera Cells Biotechnologies Co. Ltd., were randomly divided into three groups: experimental group A, group B and control group C, with 4 monkeys in each group. Paraquat were injected into the cynomologus monkeys in experimental groups, after which the monkeys in group A died of euthanasia after 5 month, and those in group B died after 2 month (2 monkeys were killed in control group in every stage). Immunohistochemical method and Western blotting analyses were applied to detect the expression feature and levels of α-syn in pancreas of cynomolgus monkeys. The result showed that α-syn immunoreactive substances were mainly observed in cytoplasmic of islet and there were differential expression of α-syn in different groups. The expression level of α-syn was highest in the experimental group A 5 months after paraquat-injecting, while the control group was at the lowest level. Our research indicated that α-syn expression in pancreas of cynomologus monkeys were up-regulated by intraperitoneal injection of small dose of paraquat, and paraquat was one of the key factors to affect the different expressions of α-syn. It provided new ideas for disease-model making, early diagnosis and intervention.
