摘  要:以杉木不同基因型的未成熟胚为起始外植体,研究不同冷藏时间、不同接种方式、不同基因型和不同激素浓度组合对胚性愈伤组织诱导率的影响。结果表明,冷藏显著影响种子的质量,随着冷藏时间延长,坏死种子比例显著增加,冷藏20 d时坏死种子比例达54.5%;冷藏对胚性愈伤组织诱导率无显著影响。接种方式对胚性愈伤组织诱导具有显著影响,接种方式二(剥去种皮,切开小口)的诱导效果最好,方式一(种子切开小口)和方式三(剥出合子胚)不利于胚性愈伤组织诱导。不同基因型在同一激素组合条件下的胚性愈伤组织诱导率存在显著差异,同一基因型在不同激素组合下的胚性愈伤组织诱导率也存在显著差异。总体看来,6-BA对胚性愈伤组织诱导率的影响最大,其最优浓度为1.0 mg/L;其次为2,4-D和ABA,其较优浓度均为2.0 mg/L;KT较优浓度为1.0 mg/L。Immature zygotic embryos of different genotypes of Chinese fir were used as initial explant in this study to explore the effects of different cold storage periods, different inoculation ways, different genotypes and different hormone combinations on embryogenic callus induction. The results showed that cold storage had signifi- cant effect on seed quality. The ratio of bad seed increased with the extension of cold storage time, which reached 54.5% after twenty days' cold storage. However, cold storage had no significant effect on the ratio of embryogenic callus induction. On the contrary, there were significant effects of different inoculation ways on the ratio of em- bryogenic callus induction. The second inoculation way (peeling and curing a small hole on end of seed) had the best effect, but both the first (curing a small hole on end of seed) and the third (taking out zygotic embryo from en- dosperm) inoculation way were not suitable for embryogenic callus induction. Different genotypes had different ra- tios of embryogenic callus induction on the same hormone combination, and the same genotype also had different ratios ofembryogenic callus induction on different hormone combinations. Overall, 6-BA had the greatest effect on the ratio of embryogenic callus induction, and its best concentration was 1.0 mg/L, and both 2,4-D and ABA was the second effective with the better concentration of 2.0 mg/L, and the better concentration of KT was 1.0 mg/L.
