摘  要:目的探讨输注不同浓度聚合人胎盘血红蛋白(Poly PHb)对大鼠造成的氧化损伤。方法建立动物换血模型,将50只SD大鼠随机均分为假手术组:仅做股动/静脉插管处理;氯化钠注射液(对照)组:股动/静脉插管,输注氯化钠注射液行换血实验;2%、4%及6%Poly PHb 3个实验组:股动/静脉插管,分别输注相应浓度的Poly PHb行换血实验。分别在大鼠换血前、换血70%后即刻观测平均动脉压(MAP),换血完成后即刻从大鼠股动脉取血0.5 m L/只,做动脉血血气分析,并检测各组大鼠血浆中还原型谷胱甘肽(GSH)浓度、超氧化物歧化酶(SOD)活力、总抗氧化能力(T-AOC)、丙二醛(MDA)浓度、8-羟基脱氧鸟苷(8-OHDG)浓度。结果 1)大鼠换血70%后,2%、4%及6%Poly PHb组与对照组相比,GSH(μmol/L)分别为33.76±14.33、47.98±21.88、39.73±15.54及10.25±5.70(P<0.01);SOD(U/m L)分别为238.07±27.39、355.73±35.42、405.26±45.01及138.46±57.16(P<0.01);T-AOC(U/m L)分别为20.50±7.87、32.65±4.38、38.03±4.50及5.40±2.22(P<0.01)。2)大鼠换血70%后,2%、4%及6%Poly PHb组与对照组相比,MDA浓度(nmol/m L)随着Poly PHb浓度的增高而增高:45.08±4.12、54.39±3.16、58.52±6.33及2.85±0.59(P<0.01)。3)大鼠换血70%后,2%、4%及6%Poly PHb组与对照组相比,大鼠血浆8-OHDG浓度(nmol/m L)未发生明显变化:2.54!0.90、2.76!0.99、2.79!0.82及2.26!0.42(P>0.05)。结论 Poly PHb对大鼠造成脂质过氧化损伤,且氧化损伤程度随着Poly PHb浓度的增高而加重,但并未对大鼠DNA造成损伤。Objective To explore the oxidative damage caused by different concentration of polymerized human placenta hemoglobin (PolyPHb) in rats. Methods The rat blood replacement model was established. 50 SD rats were randomly divided into placebo group: femoral artery/vein intubation ; NaCl (control) group (femoral artery/vein intubation and blood replacement experiment) ; experimental group (2% ,4%, 6% PolyPHb) (femoral artery/vein intubation and blood replacement experiment) MAP and arterial blood gas before the rat blood replacement and the end of the blood replacement were monitored and measured. After finishing the blood replacement, arterial blood and separate plasma were taken to detect reduced glutathione (GSH) concentration, superoxide dismutase (SOD) activity, total antioxidant capacity (T-AOC) , malondialdehyde (MDA) concentration, 8-hydroxy-2 deoxyguanosine (8-OHDG) concentration. Results After 70% blood replacement, different concentrations of PolyPHb in different groups were compared. GSH concentration, SOD activity and T-AOC were significantly increased, and the results were (1) GSH(μmol/L) :33.76± 14. 33 vs 47.98±21.88 vs 39. 73± 15.54 vs 10. 25±5.70(P〈0. 01 ) ; (2) SOD(U/mL) : 238.07±27.39 vs 355.73±35.42 vs 405.26±45.01 vs 138.46±57. 16( P〈 0.01 ) ; (3) T-AOC(U/mL) :20. 50±7. 87 vs 32. 65±4. 38 vs 38.03±4. 50 vs 5.40±2. 22( P〈0. 01 ). The concentration of MDA(nmol/mL) increased with the concentration of PolyPHb, and the results were 45.08±4. 12 vs 54. 39±3.16 vs 58.52± 6. 33 vs 2. 85±0. 59(P〈0. 01 ). The 8-OHDG concentration (ng/mL) did not have obvious change, and the resuhs were: 2. 54±0. 90 vs 2.76±0. 99 vs 2.79±0. 82 vs 2. 26±0.42(P〉0.05). Conclusion PolyPHb induces lipid peroxidation in rats, and the degree of oxidative damage increases with the increase of PolyPHb concentration. In addition, PolyPHb does not cause DNA damage in rats.
