摘 要:目的探讨高糖对肾小球系膜细胞细胞外基质表达的影响,以及西格列汀(SIT)对其的干预作用。方法培养肾小球系膜细胞,分为对照(Con)组、高糖(HG)组,以及高糖联合不同浓度的SIT组(HG+S1组、HG+S5组、HG+S10组和HG+S20组)。培养24 h、48 h后,用MTT法测定细胞增殖程度,ELISA检测细胞培养上清层粘连蛋白(LN)、大鼠Ⅳ型胶原蛋白(Col-Ⅳ)、转化生长因子β1(TGF-β1)蛋白表达的变化。结果高糖培养24 h、48 h后,可观察到不同浓度的SIT均可抑制高糖诱导的系膜细胞增殖;SIT组与HG组比较,差异有统计学意义;Con组肾小球系膜细胞可分泌一定量的LN、Col-Ⅳ和TGF-β1,高糖刺激后LN、Col-Ⅳ和TGF-β1分泌增加,而SIT对系膜细胞分泌物的抑制随其浓度的升高而增强。结论SIT可抑制高糖条件下系膜细胞的增殖,且能降低高糖条件下LN、Col-Ⅳ、TGF-β1的表达,SIT可能对糖尿病患者的肾脏有一定的保护作用。Objective To investigate the interventional effect of Sitagliptin (SIT) on secretion of extracellular matrix of glomerular mesangial cells cultured in high glucose ambient (HG). Methods The glomerular mesangial cells were cultured and divided into six groups according to different intervention: Control group (Con), HG group, HG+SI group, HG+S5 group, HG+S10 group, HG+S20 group. After cultured for 24 h or 48 h, the cell proliferation conditions were detect by MTT method. The protein expressions of LN, Col-Ⅳ and TGF-β1 in the supernatant were measured by ELISA method. Results The proliferative effect of high glucose on the cultured mesangial cells was significantly stimulated in 24 h and 48 h incubation phases. Compared with HG group, SIT with different concentrations could significantly inhibit the HG induced cell proliferation (P〈0. 05). The glomerular mesangial cells in Con group could secret a certain amount of LN, Col-Ⅳ and TGF-β1, which were all stimulated by HG. The inhibition effect of sitagliptin on mesangial cells was increased together with the increase of its concentration in a doses -dependent manner. Conclusion SIT can inhibit the HG induced cell proliferation,and lower the expression of LN, Col-Ⅳ and TGF -6 1 in HG ambient, SIT may have a protective effect on kidney in diabetic patients.
