摘 要:目的探讨百草枯(paraquat,PQ)对星形胶质细胞合成与释放的非递质物质水平的影响。方法人星形胶质(HA1800)细胞分别暴露于终浓度为0、200、400和800μmol/L的PQ 12、24和48 h,ELISA试剂盒法检测细胞培养上清液中胶质源性神经营养因子(GDNF)和脑源性神经营养因子(BDNF)水平。胆固醇定量检测试剂盒法和Western Blot法分别检测0和400μmol/L的PQ对细胞膜中胆固醇含量和载脂蛋白E(Apo E)表达水平的变化。结果与相应时点的阴性对照组比较,PQ能显著降低细胞膜中胆固醇含量和Apo E表达水平(P<0.05),PQ也能以剂量依赖方式降低细胞培养上清液中GDNF和BDNF水平(P<0.05)。结论 PQ能通过影响星形胶质细胞非递质类物质水平,参与调控突触发生及可塑性,进而参与神经退行性疾病的发生。Objective To investigate the effect of paraquat on the synthesis and release of non-neurotransmitters in the astrocytes. Methods Human astrocyte (HA1800) cells were exposed to a final concentrations of 0, 200, 400 and 800 Ixmol/L PQ for 12, 24 and 48 h, The levels of glial cell line-derived neurotrophic factor (GDNF) and brain-derived neurotrophic factor (BDNF) in the culture supernatants were measured by ELISA kit. Cholesterol content and apolipoprotein E (ApoE) expression in the cell membrane were detected by Cholesterol Quantitative Assay Kit and Western Blot assay respectively at 0 and 400 p, mol/L PQ groups. Results Compared with the negative control group at corresponding time point, PQ could significantly reduce the level of cholesterol content and the expression of ApoE in cell membranes ( P 〈 0. 05 ) , PQ also showed a dose-dependent decrease in cell culture supernatant of GDNF and BDNF ( P 〈 0. 05 ). Conclusions PQ can be involved in the regulation of synapse formation and plasticity by affecting the level of non neurotransmitter substance in astrocytes, and then participate in the occurrence of neurodegenerative diseases.
