摘  要:目的应用RNA干扰抑制基质金属蛋白酶-3(matrix metalloproteinase-3,MMP-3)基因的表达,探讨MMMP-3基因抑制后对血管平滑肌细胞(Vascular Smooth Muscle Cells,VSMCs)迁移的影响。方法采用离体培养大鼠主动脉平滑肌细胞,设计、合成靶向大鼠MMP-3基因的小干扰RNA序列,筛选有效的干扰序列,用瞬时转染法转染VSMCs。将血管平滑肌细胞培养24 h后随机分为:对照组、Lipofectamine 2000组、血小板源生长因子(PDGF)组、PDGF+MMP-3-siRNA-1组及MMP-3-siRNA-1组;用siRNA体外转染体外培养的血管平滑肌细胞,采用改良的Boyden微孔膜双槽法进行细胞迁移实验。结果 MMP-3-siRNA-1能够显着抑制PDGF诱导的VSMCs细胞迁移,抑制率为32.1%。结论 siRNA干扰介导MMP-3基因沉默可抑制血管平滑肌细胞的迁移。MMP-3基因可能成为治疗心血管疾病一个新的靶点。Objective To explore the effect of silencing MMP-3 gene with RNA interference (RNAi) on the migrationof vascular smooth muscle cells ( VSMCs). Methods Based on SHR MMP-3gene sequence and RNAi sequence designprinciples ,MMP-3 small interfering RNA (siRNA) were designed and synthesized, the most effective target sequencewas chosen. VSMCs were divided into five groups randomly. Control group, Lipofeetamine 2000 group, PDGF group,PDGF+MMP-3-siRNA-lgroup and MMP-3-siRNA-1 group. Modified Boyden chamber technique was employed forVSMCs migration assessment. Results RNAi targeting MMP-3 gene significantly inhibited. The migration of VSMCsinduced by PDGF was significantly inhibited by MMP-3 -siRNA -1 were at 32.1% (P〈O.01).Conclusions RNAitargeting at MMP-3 may be a potential approach for inhibition of cell proliferation and migration involved inpathogenesis of proliferative vascular diseases.
