摘 要:目的:应用微量热法检测两种药物对白色念珠菌的抑制效果。方法:挑取白色念珠菌单菌落接种于沙氏培养基中,37℃恒温摇床培养,然后将菌液稀释至108 CFU/ml备用。再将稀释后的细胞贮备液、培养基及不同浓度的药物加入安瓿瓶中,密封、擦净安瓿瓶后将其放入TAM量热仪测量通道中,记录反应过程的热输出功率曲线。结果:获得了白色念珠菌在不同药物作用下的热谱曲线,这两种药物对白色念珠菌的生长都有明显抑制作用。结论:根据药物浓度与抑制率的关系求出了两种药物的半抑制浓度,说明抑菌效果氯已定强于益康唑。氯已定的有效杀菌浓度为5.3μmol/L。Objective: To investigate the inhibitory effects of two drugs by micro calorimetry on candida albicans. Methods: The candida albicans were cultivated in Salouraud Liquid Medium at 37 ℃. Then bacteria solution was collected from the medium and diluted to 108 CFU/ml(CFU,ColonyForming Units) as stock solutions. Furthermore, stock solutions, Salouraud Liquid Medium and test drugs with different concentrations were mixed in the ampoule, then, the ampoules were put in TAM air, and the thermal variation curves of reaction process were obtained. Results: The effective bactericidal concentration of chlorhexidine was 5.3 μmol/L. According to the metabolic heat spectrogram, we found that both chlorhexidine and econazole were potential good antifungal drugs. Conclusion: The order of inhibitory effect of the two drugs is chlorhexidine〉econazole.
