长链非编码RNA GAS5在子宫内膜癌中的表达及对肿瘤侵袭的影响

摘　　要：目的探讨长链非编码RNA GAS5在子宫内膜癌组织中的表达及其对子宫内膜癌HEC-1A细胞侵袭能力的影响。方法筛选2013年1月-2015年12月间于解放军451医院妇产科行手术切除的子宫内膜癌患者的癌及癌旁组织50例,分析lncRNA-GAS5在组织中的表达水平及临床意义;通过重组质粒在人子宫内膜癌HEC-1A细胞中过表达lncRNA-GAS5,采用Transwell侵袭小室模型检测过表达lncRNA-GAS5后HEC-1A细胞侵袭能力变化,qRT-PCR检测micro RNA-21的表达变化,qRT-PCR及Western blot检测PTEN表达变化。结果 lncRNA-GAS5在子宫内膜癌组织中表达水平低于对应癌旁组织(t=4.013,P<0.05),子宫内膜癌组织中长链非编码核醣核酸生长组织特异性转录体5(lncRNA-GAS5)低表达与肿瘤淋巴结转移(t=2.389,P=0.017)及高FIGO分期(Ⅲ+Ⅳ期,t=2.131,P=0.021)差异有统计学意义;lncRNA-GAS5重组质粒转染HEC-1A细胞后可抑制细胞的侵袭能力(t=7.654,P=0.008);qRT-PCR检测发现过表达lncRNA-GAS5后microRNA-21(miR-21)的表达水平升高(t=23.533,P<0.05),qRT-PCR及Western blot结果证实,过表达lncRNA-GAS5后可促进PTEN的表达(P<0.05)。结论 lncRNA-GAS5在子宫内膜癌组织中表达降低,lncRNA-GAS5可能通过下调miR-21的表达进而恢复PTEN的表达来抑制子宫内膜癌转移。Objective To study the expression of long noncoding RNA GAS5 （lncRNA-GAS5） in endometrial carcinoma and its role in regulation of cell invasion. Methods Endometrial carcinomas and the matched tumor adjacent tissues were collected from 50 cases who received surgical treatment in our hospital between January 2013 and December 2015. The expression of lneRNA-GAS5 was detected by qRT-PCR. LncRNA-GAS5 over-expression plasmid was transfected into HEC-1A cells. Transwell invasion assay model was used to measure cell invasion. The expressions of miR-21 and PTEN were detected by qRT-PCR and Western blot. Results The expression of lneRNA-GAS5 in the endometrial carcinoma tissues was significantly lower than that in the adjacent tissues （t = 4.013, P 〈 0.05）. Low expression of lneRNA-GAS5 was associated with lymphatic metastasis （t = 2.389, P = 0.017） and advanced FIGO stage （stage III and IV; t = 2.131, P = 0.021）. Over-expression of lncRNA-GAS5 could significantly suppress the invasive ability of HEC-1A cells （t = 7.654, P = 0.008）. Moreover, over-expression of lncRNA-GAS5 promoted miR-21 and PTEN expressions （P 〈 0.05）. Conclusions lncRNA-GAS5 expression is lowerd in endometrial carcinoma tissues and associated with cell invasion. LncRNA-GAS5 may suppress metastasis of endometrial carcinoma by inhibiting miR-21/PTEN axis.
