摘  要:为了拯救濒危植物——北苍术,保护种质资源,在山西省中条山和太行山采集北苍术,以北苍术的种子为材料获得无菌试管苗,分别利用根、胚轴、叶片为外植体,研究了外植体对愈伤组织诱导率的影响;应用正交试验法研究6-卡基腺嘌呤(6-BA)、激动素(KT)、萘乙酸(NAA)及其配比对北苍术无菌苗增殖的影响;同时研究了NAA浓度对北苍术生根的影响。结果表明:以胚轴为外植体在1/2MS+2,4-D2.0 mg/L+KT 1 mg/L+蔗糖3%的培养基中更有利于形成愈伤组织从而长成完整植株;北苍术的最适增殖培养基为:1/2MS+6-BA 2 mg/L+KT 1 mg/L+NAA 0.5 mg/L+蔗糖3%;最适生根培养基为:1/2MS+NAA 0.5 mg/L+蔗糖3%。To save the endangered plant—— Atractylodes chinensis, and protect germplasm resources, Atractylodes chinensis was collected in Zhongtiao Mountain and Taihang Mountain in Shanxi Province. Sterile test tube seedling was obtained from the seeds of Atractylodes chinensis. Taking root, hypocotyl and leaf as explants, the effects of explants on the induction rate of callus was studied. The effects of 6-BA, KT, NAA and their proportioning on the proliferation of Atractylodes chinensis sterile seedling were studied with orthogonal test method. The effect of NAA on the rooting of Atractylodes chinensis was also analyzed. The results showed that hypocotyl as the explants was easy to produce the callus in the medium 1/2MS+2,4-D 2.0 mg/L+KT 1 rag/L+ sugar 3%, and then developed into the complete plant. The optimum medium of proliferation was 1/2MS+6-BA 2 mg/L+KT 1 mg/L+NAA 0.5 mg/L+ sugar 3%. The optimum medium of rooting was 1/2MS+NAA 0.5 mg/L+ sugar 3%.
