摘 要:目的观察宁夏枸杞总黄酮(total flavonoids from lycium barbarum,TFL)对MCF-7细胞株细胞增殖和凋亡的影响。方法从枸杞中提取TFL,采用细胞增殖与细胞毒性检测试剂(Cell Counting Kit 8,CCK8)法观察不同TFL作用浓度(30、75、150μg·mL^(-1))对MCF-7细胞增殖的抑制作用;流式细胞术检测TFL对MCF-7细胞凋亡的影响;反转录PCR法检测TFL对MCF-7细胞Bax、Bcl-2、Caspase-3的m RNA表达的影响。结果 TFL作用MCF-7细胞48h的半数抑制浓度(half maximal inhibitory concentration,IC50)为75.58μg·mL^(-1);TFL抑制MCF-7细胞的增殖,且随作用浓度增加,其抑制作用增强(P均<0.05);在药物作用48h时的抑制作用较24h时强(P<0.05)。流式细胞术显示20、37.5μg·mL^(-1)的TFL可促进MCF-7细胞凋亡作用,且随浓度增加,其凋亡率增加;20、37.5μg·mL^(-1)的TFL作用于MCF-7细胞,MCF-7细胞中Caspase-3表达随浓度增加逐渐升高,Bcl-2表达随浓度增加逐渐下降(P<0.05)。结论 TFL可抑制乳腺癌MCF-7细胞增殖,促进其凋亡,为进一步研究TFL在乳腺癌防治中的作用提供了实验依据。Objective To investigate the effects of total flavonoids fromLycium barbarum(TFL)on proliferation and apoptosis of human breast cancer cell line MCF-7. Methods The proliferation inhibition rate of MCF-7cells was measured by cell counting kit-8 assay. Apoptosis rate of MCF-7 cells was detected with Annexin V-fluorescein isothiocyanate/propidium iodide double staining by flow cytometry. The expressions of Bax,Bcl-2and caspase-3 were analyzed by RT-PCR. Results When the concentration of TFL were 30,75,150μg·mL^(-1),TFL could inhibit the proliferation of MCF-7 cells,and the inhibitory effect increased with concentration increasing. At the 24^(th) h and 48^(th) h,the inhibitory effect of drugs on the cells was increased with the prolongation of action time. The half maximal inhibitory concentration(IC50)was 75.58μg·mL^(-1). The result of flow cytometry showed that 20,37.5μg·mL^(-1)TFL could promote the apoptosis of MCF-7 cells,and the apoptosis rate increased with the increase of concentration. When the concentration of TFL was 20 and 37.5μg·mL^(-1)in MCF-7 cells,the expression of Bax and Caspase-3 in MCF-7 cells increased gradually,and the expression of Bcl-2 decreased gradually with the increase of TFL concentration. Conclusion TFL can inhibit the proliferation and promote the apoptosis of breast cancer MCF-7 cells. This study will provide experimental evidence for the further research of the effect of total flavonoids fromLycium barbarum in the prevention and treatment of breast cancer.
