摘  要:目的:研究活血利湿方对非酒精性脂肪性肝病(Non-alcoholic fatty liver disease,NAFLD)大鼠肝功能及肝脏病理的影响。方法:利用高脂饲料建立SD大鼠非酒精性脂肪肝模型,期间预防组大鼠予活血利湿方灌胃,8周造模成功后,取血及肝脏,观察活血利湿方的预防效果。造模成功大鼠分为三组,分别予活血利湿方水煎剂、荷丹片溶液、生理盐水4周后,取血及肝脏。两批大鼠均检测血清中谷丙转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)水平,同时观察大鼠肝组织病理学形态的变化,评估NAFLD活动度积分(NAFLD activity score,NAS)及炎症分级。结果:与空白组相比,模型组大鼠ALT水平不变(P>0.05),AST、LDH升高(P<0.05),预防组AST、LDH较模型组降低。与模型组相比,活血利湿方组NAS总积分及炎症分级均下降(P<0.05),有统计学差异。结论:活血利湿方可改善非酒精性脂肪肝大鼠肝功能、肝脏病理形态,减轻炎症程度。Abstract: Objective: To study the effect of Promote-blood-circulation and Remove-dampness Decoction (PARD) on liver function and pathological changes in SD rats with non-alcoholic fatty liver disease (NAFLD). Methods: 8 rats fed with general di- et were taken as blank controlled group. To induce NAFLD, a high-fat diet was given to SD rats of prevention group (n = 8) and model group ( n = 27 ). During this period, the rats in prevention group were fed PARD by gastric perfusion. After 8 weeks, 2 rats were selected from model group randomly and were killed to decide the animal model was successful by observing pathological changes in the liver. Rats in the blank controlled group, the prevention group and the model group (7 of them) were executed to obtain the plasma and liver tissues. All the liver tissues were stained with Hematoxylin-Eosin (HE) , the tissue of rats in the rood-el group were histologically analyzed and the animal model was successful. The effect of PARD on serum levels of alanine amin- otransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH) were detected during the period of model building. Meanwhile, all the liver tissues were observed to learn pathological changes. Results- The levels of ALT had no differ- ence between the blank controlled group and the model group (P 〉 0. 05). Compared with the blank controlled group, the levels of AST, LDH in the model group were rise (P 〈 0. 05). The prevention group had lower (P 〈0.05) levels of AST, LDH com- pared with the model group. NAS score and inflammatory status were all reduced in the PARD' group ( P 〈 0. 05), compared with the model group. Conclusion: PARD can improve liver function, liver pathological morphology and reduce the degree of he- patic inflammation of rats with NAFLD.
