摘 要:目的:观察芪明颗粒对DN大鼠模型大鼠肾组织中WT1、AngⅡ、ET-1表达的影响,研究其对DN的作用机制。方法:选用SPF级SD大鼠50只,除空白组外各组给予链脲佐菌素单次腹腔注射造模,随机分为空白组、模型组、低剂量组、高剂量组、缬沙坦组。8周后分别用Westernblot法、ELISA法检测肾组织中WT1、ET-1、AngⅡ的表达。结果:模型组肾组织WT1、AngⅡ、ET-1明显高于空白对照组(P<0.05);与模型组相比,低剂量组、高剂量组、缬沙坦组肾脏组织WT1、AngⅡ、ET-1均显著降低(P<0.05);高剂量组、缬沙坦组肾组织WT1、AngⅡ、ET-1明显低于中药低剂量组(P<0.05),中药高剂量组、缬沙坦组相比差异无统计学意义(P>0.05)。结论:芪明颗粒可以有效的降低DN大鼠肾组织WT1、AngⅡ、ET-1的表达,以起到保护肾脏的作用。高剂量芪明颗粒较低剂量对肾脏有更好的保护作用。早期采用芪明颗粒对糖尿病进行干预,可以延缓肾损害的进展,为临床广泛应用芪明颗粒提供实验基础和理论依据。Objective: To observe the effect of Qiming Granule on the expression of WT1, Ang and ET-1 in renal tissue of DN rats, and to study the mechanism of DN. Methods: 50 SD grade SPF rats were selected and randomly divided into blank group, model group, low dose group, high dose group and valsartan group. After 8 weeks, the expression of WT1, ET-1 and Ang in renal tissues were detected by Westernblot and ELISA methods respectively. Results : The renal tissue of WT1, AngⅡ, ET-1 in the model group was significantly higher than the controlled group ( P 〈 0. 05 ) ; compared with the model group, kidney tissue WT1, AngⅡ and ET-1 of the low dose group, the high dose group, the valsartan group were significantly decreased (P 〈 0. 05 ) ; renal WT1, Ang Ⅱ, ET-1 of the high dose group, the valsartan group of was significantly lower than that of the low dose group ( P 〈 0.05 ) , there was no significant difference ( P 〉 0. 05 ) between the high dose group and valsartan group. Conclusion : Qiming Granule can effectively reduce the expression of WT1, Ang and ET-1 in renal tissue of DN rats, which can protect the kidney. High dose of Qiming Granule has better protective effect on kidney. In the early stage, it can delay the progression of renal dam- age by using Qiming Granule to interfere with diabetes. To provide experimental basis and theoretical basis for the clinical appli- cation of Oiming Granule.
