加减血癥汤对人胃癌细胞SGC-7901增殖、凋亡及周期的影响

摘　　要：目的:探讨加减血癥汤对人胃癌细胞SGC-7901增殖、凋亡及细胞周期的影响。方法:采用甲基噻唑(MTT)比色法和生长曲线测定不同浓度加减血癥汤在不同作用时间内对在体外培养的SGC-7901细胞增殖的影响,同时应用流式细胞术检测凋亡率及细胞增殖周期的变化。结果:加减血癥汤可明显抑制SGC-7901细胞的增殖,并呈时间-剂量依赖性;相同作用时间下,加减血癥汤的浓度越高,对肿瘤细胞生长的抑制作用越强,凋亡率也越高,处于G0/G1期的细胞比例逐渐增多,S期细胞比例出现相对减少。结论:加减血癥汤能抑制人胃癌细胞SGC-7901生长,诱导细胞凋亡,并使细胞周期阻滞于G0-G1期。Objective： To investigate the suppressive effect of Modified Xuezheng Decoction on the growth and apoptosisthe and cell cycle of human gastric carcinoma cell line SGC-7901. Methods ： The survival rate and the inhibitory rate of SGC-7901 cell in vitro were detected by MTF colorimetrie assay and cell growth curve assay at different time points under different concentra- tion of modified Xuezbeng Decoction. The apoptotie rate and cell cycle were investigated by flow cytometryanalysis. Results ： The Modified Xuezheng Decoction could obviously inhibit the SGC-7901 cell proliferation in a dose and time-dependent manner. It raised the apoptoticrate in a concentration depending manner in a certain time, while the cells of GO/G1 phase increased, the cells of S phase decreased relatively. Conclusion： Modified Xuezheng Decoction can suppress the proliferation of SGC-7901, in- duce apoptosis and block cell cycle at G0/G1 phase.
