摘 要:目的基于"肝藏"理论,通过对肝郁血虚ED大鼠的观察,探讨"疏肝养血法"调控阴茎海绵体平滑肌的物质基础与效应机制。方法将30只SD大鼠随机分3组,即空白组、模型组、治疗组,每组各10只。治疗组予以四物汤加逍遥散合方治疗,空白组、模型组给予等体积生理盐水,连续14d。观察大鼠性活动情况、阴茎海绵体平滑肌形态及钙离子通道蛋白CaV1.3、RyR1的表达。结果(1)与空白组比较,模型组大鼠性活动能力明显减弱,即骑跨潜伏期[(158.16±27.42)s]、插入潜伏期[(525.23±105.56)s]延长,骑跨次数[35.31±6.17]与插入次数[8.84±1.48]明显减少(P<0.05);与模型组比较,治疗组骑跨潜伏期[(96.07±18.26)s]、插入潜伏期[(310.27±104.24)s]缩短,骑跨次数[49.6±18.65]与插入次数[15.21±4.63]显著改善(P<0.05)。(2)各组阴茎海绵体平滑肌病理切片HE染色后,形态学改变不明显(P>0.05)。(3)与空白组比较,模型组大鼠阴茎海绵体平滑肌CaV1.3[22059.51±10945.64]及RyR1蛋白[25053.70±2941.02]表达显著降低(P<0.05);与模型组比较,治疗组CaV1.3[42386.31±9515.26]及RyR1蛋白[38038.25±4980.82]表达显著改善(P<0.05)。结论疏肝养血法可显著改善肝郁血虚ED大鼠阴茎组织Ca2+通道蛋白CaV1.3及RyR1的表达,可能是"肝藏"的理论物质基础及效应机制的重要环节。Objective To investigate the material basis and effective mechanisms"Dispersing liver and Nourishing blood" regulates the corpus cavernosum smooth muscle based on the guidance of theory"Liver Storing". Methods Thirty SD rats were randomly divided into three groups: blank group, model group, treatment group, 10 rats in each group. The treatment group was treated with Siwu Decoction with Xiaoyao Powder. Blank group, model group given equal volume of saline continuous treatment for 14 days. During the experiment, the sexual activity of the rats, the smooth muscle morphology of the corpus cavernosum and the expression of CaV1.3 and RyR1 were observed. Results 1. Compared with that in blank group, the rats in model group had lower sexual activity with longer cross-latency[158.16±27.42s] and intromission latency[525.23±105.56s], lower frequency of straddling[35.31±6.17] and insertions [8.84±1.48] (P〈0.05).Compared with those of the model group, across latency [49.62±18.65], insert latency [310.27±104.24], straddling times [96.07±18.26] and the number of insertions [15.21±4.63] of rats in the treatment group were significantly improved (P〈0.05). 2. No obvious changes in morphology of corpora cavernosa tissue were found (P〉0.05). Compared with that of the control group, the expressions of CaV1.3 [22059.51±10945.64] and RyR1 protein [25053.70±2941.02] in rat corpus cavernosum smooth muscle was significantly downregulated in the model group (P〈0.05); Compared with that of the model group, the expressions of CaV1.3 [42386.31±9515.26] and RyR1 protein [38038.25±4980.82] was significantly upregulated in the treatment group (P 〈0.05). Conclusion"Dispersing liver and Nourishing blood" could significantly increase the expressions of CaV1.3 and RyR1 and promote Ca2+ outflowing, ultimately result in relax the penile smooth muscle, which may be one of material basis of theory"Liver Storing".
