摘 要:目的观察益智宁神颗粒对幼年自发性高血压大鼠(SHR)活动水平和空间记忆能力的影响。方法将64只SHR随机分为模型组、中药组、托莫西汀组、哌甲酯组,每组16只。将16只京都大鼠作为正常组。中药组给予益智宁神颗粒1.1 g/(kg·d)灌胃,托莫西汀组给予托莫西汀胶囊3 mg/(kg·d)灌胃,哌甲酯组给予哌甲酯片5 mg/(kg·d)灌胃,模型组和正常组给予与中药组等体积的生理盐水灌胃。每天1次,连续4周。通过开场实验和水迷宫实验观察大鼠的活动水平及认知功能。结果开场实验:模型组大鼠直立次数和水平移动格数明显大于正常组(P<0.05);中药组大鼠直立次数明显少于模型组、哌甲酯组及托莫西汀组,但仍高于正常组(P<0.05)。模型组、中药组、哌甲酯组及托莫西汀组间水平移动格数比较差异无统计学意义(P>0.05)。Morris水迷宫实验:定位航行实验部分,随着训练时间的增加,中药组、托莫西汀组、哌甲酯组潜伏期呈现平缓下降趋势。与正常组各时间点比较,模型组、中药组、托莫西汀组、哌甲酯组潜伏期均缩短(P<0.05)。大鼠游泳速度、游泳距离随训练时间的增加未出现明显改变。空间探索实验部分,与正常组比较,模型组、中药组、托莫西汀组、哌甲酯组潜伏期缩短,托莫西汀组游泳速度增加(P<0.05);各组大鼠间游泳距离差异无统计学意义(P>0.05)。结论益智宁神颗粒可以改善幼年SHR冲动、多动行为及认知水平,疗效与托莫西汀及哌甲酯相似,远期疗效优于托莫西汀及哌甲酯。Objective To observe the impact of Yizhi Ningshen Keli (益智宁神颗粒) on the activity level and spatial memory ability of juvenile spontaneous hypertension rats (SHR). Methods Sixty-four SHR were random- ized into model group, Chinese medicine group, tomoxetine group and methylphenidate group, each group with 16 rats. Sixteen Kyoto rats were taken as the normal group. The Chinese medicine group was given Yizhi Ningshen Keli 1.1 g/(kg · d) by gavage. The tomoxetine group was given tomoxetine capsule 3 rag/( kg · d) by gavage. The methylphenidate group was given methylphenidate tablet 5 mg/( kg · d) by gavage. The model group and the normal group were given normal saline equivalent to the Chinese medicine group by gavage, once every day, for 4 weeks. Rats' activity level and cognitive function were observed by open field test and water maze test. Results In the open field test, rats' erect times and horizontally moving grids number in the model group were larger than those in the normal group (P 〈 0.05 ). Rats' erect times in the Chinese medicine group were smaller than those in the model group, the methylphenidate group and the tomoxetine group, while the times were larger than in the normal group ( P 〈 0.05 ). The difference of horizontally moving grids number in the model group, the Chinese medicine group, the methylphenidate group and the tomoxetine group was not significant (P 〉 0. 05 ). In the Morris water maze test, as for place navigation test part, with the increase of test time, the latent period showed a gentle downward trend in the Chinese medicine group, the tomoxetine group and the methylphenidate group. Compared each time point with the normal group, the latent period decreased in the model group, the Chinese medicine group, the tomoxetine group and the methylphenidate group (P 〈0.05 ).
