摘  要:目的探讨针刺治疗血管性痴呆的可能作用机制。方法 24只大鼠随机分为假手术组、模型组、针刺组和非穴组,每组6只。除假手术组外其余各组应用双侧颈总动脉永久性结扎方法制备血管性痴呆大鼠模型。应用Nissl染色观测其皮质神经元形态学改变,应用TUNEL法观察皮质神经元凋亡情况,应用免疫荧光技术观察磷酸化c-Jun氨基末端激酶(p-JNK)表达。结果与假手术组比较,模型组大鼠大脑皮质细胞排列散乱,核固缩、深染,且大脑皮质凋亡神经元增多,p-JNK阳性细胞数增多(P<0.05);与模型组比较,针刺组大鼠皮质神经元形态改善,凋亡神经元较模型组显著减少,p-JNK表达减少(P<0.05);而非穴组无明显改善。结论针刺可抑制血管性痴呆大鼠大脑皮质p-JNK表达,减少大脑皮质凋亡神经元,可能是其治疗血管性痴呆的机制之一。Objective To explore the possible mechanism of acupuncture treating vascular dementia (VD). Methods Twenty-four rats were randomized into sham operation group, model group, acupuncture group and non acupoint group with 6 rats in each group. Except the sham operation group rats in other groups were built VD rat models using permanent bilateral common carotid artery ligation method. Cortical neuron morphological changes were examined by Nissl stain. Cortical neuron apoptosis were examined by TUNEL method. The expression of phosphoryla- tion c-Jun NH2-terminal Kinase (p-JNK) was examined by immunofluorescence technique. Results Compared with the sham operation group, the model group rats' cortical cells were scattered, nucleus pyknosis and deep staining. Cortical neuron apoptosis and p-JNK expression increased (P 〈 0. 05 ). Compared with the model group, the acupuncture group rats' cortical neuron morphology improved. Neuron apoptosis and p-JNK expression decreased (P 〈 0.05 ). There was no obvious improvement in the non acupoint group. Conclusion Acupuncture might inhibit the expression of p-JNK in cortex of VD rats. Reducing cortical neuron apoptosis might be one of the mechanisms of treating VD.
