摘  要:目的观察升降散对糖尿病大鼠乳酸杆菌、肠链球菌及TNF-α的影响,探讨其作用机制。方法链脲佐菌素诱导建立糖尿病模型,成模大鼠分为模型对照组和治疗组(拜糖平组、升降散低剂量组、升降散中剂量组、升降散高剂量组),各组按相应药物剂量灌胃治疗6周后,采用Elisa法检测TNF-α、INS,Realtime PCR技术进行细菌定量测定。结果治疗前,模型组空腹血糖(FBG)、TNF-α、INS及肠链球菌均明显升高,乳酸杆菌数量明显减少;治疗前后,正常对照组及模型对照组各指标无明显变化。治疗后,各治疗组FBG在2周、4周及6周逐渐降低,且均高于同时间正常对照组,低于同时间模型对照组(P<0.05);TNF-α、INS较模型对照组明显减少,但同时高于正常对照组(P<0.05);肠链球菌数量较模型对照组明显减少,较正常对照组以拜糖平组及升降散中剂量组减少明显,差异均有统计学意义(P<0.05);乳酸杆菌数量较模型对照组、正常对照组明显增多(P<0.01)。结论升降散能通过增加益生菌数量,降低致病菌数量达到调整肠道菌群结构的目的,从而抑制炎症状态,降低血糖。其部分作用机制可能是通过调整肠道菌群、抑制炎症因子而减轻胰岛素抵抗(IR),增加胰岛素利用率实现的。Objective To investigate the effects of Shengjiang powder on intestinal flora structure change and inflammation factor in DM rats, and to explore its action mechanism. Methods The model rats of DM were established by injecting streptozotosin, and then they were randomly divided into model control group and treatment group (acarbose group, groups of low, medium, high dose of Shengjiang powder) . After the treatment for 6 weeks, Elisa method was used to detect TNF-a, INS, Reahime PCR technology for quantitative determination of bacteria. Results Before treatment, the FBG, TNF-α, INS and Enterococcus of the model group were significantly increased, and the lactobacillus quantity of the model group decreased significantly. The indexes of the normal control group and model control group had no significant change before and after treatment. After treatment, FBG of the treatment groups in 2 weeks, 4 weeks and 6 weeks were gradually decreased, which was higher than that of the normal control group, and which was lower than that of the model group and the control group at the same time (P 〈 0.05) . The TNF-α and INS quantity of the treatment group decreased significantly comparing with model control group, but at the same time, which was higher than that of the normal control group (P 〈 0.05) . The Enterococcus quantity of the treatment groups decreased significantly compared with model control group, but the acarbose group and medium group of Shengjiang powder decreased significantly comparing with normal control group. The lactobacillus quantity of the treatment groups increased significantly comparing with model group and normal control group (P 〈 0.01) . Conclusion Shengjiang powder can spread by increasing the number of probiotics, reducing the number of pathogenic bacteria to adjust the structure of intestinal flora, thus inhibiting the inflammatory state to reduce FBG. Some of its mechanism may be through adjusting the intestinal flora, inhibiting inflammatory factor to insulin resistance �
