纳米羟基磷灰石对正常细胞和肿瘤细胞的毒性比较研究

摘  要:目的比较纳米羟基磷灰石(HA-NPs)对肿瘤细胞和正常细胞的毒性差异,为HA-NPs的安全性提供实验证据。方法以肝肿瘤细胞(Hep G2)和肝正常细胞(L02)为研究对象,采用噻唑蓝比色(MTT)、乳酸脱氢酶(LDH)释放和台盼蓝染色三种不同的实验方法比较HA-NPs对Hep G2和L02的细胞毒性差异。结果 MTT结果显示,在HANPs染毒浓度为100、200和400μg/ml时,L02细胞存活率分别为110%、114%和104%,而Hep G2细胞存活率分别为92%、89%和73%;在HA-NPs浓度为600μg/ml^1600μg/ml时,L02细胞的存活率从90%降至27%,而Hep G2细胞的存活率则从67%降至13%,差异均有统计学意义(P<0.05)。LDH释放和台盼蓝染色结果显示,400、600和800μg/ml的HA-NPs对Hep G2和L02的细胞毒性均高于相应的对照组(P<0.05),且相同剂量的HA-NPs对Hep G2的细胞毒性要高于L02(P<0.05)。结论高浓度HA-NPs对Hep G2和L02细胞均具有毒性效应,且相同剂量的HANPs对L02的细胞毒性要低于Hep G2细胞。Objective To compare the toxicity of Nano - hydroxyapatite ( HA - NPs) on tumor cells (HepG2) and normal cells (L02) , and to provide experimental evidence for the safety of HA - NPs. Methods HepG2 and L02 cells were treated with different concentrations of HA - NPs, and the cytotoxicity of these two ceils was measured by MTT assay,lactate dehydrogenase (LDH) release assay and trypan blue staining assay. Results The results of MTT assay showed that HA - NPs exposed to the concentrations of 100,200 and 400 μg/ml, cell survival rates of L02 were 110% , 114% and 104% , and the survival rates of HepG2 cells were 92% ,89% and 73%. While the treatment concentrations of HA - NPs ranged from 600 μg/ml to 1600 trg/ ml, L02 cell survival rates decreased from 90% to 27 %, the survival rates of HepG2 cells decreased from 67% to 13%, and the differences were statistically significant ( P 〈 0. 05 ). Results from LDH release and trypan blue staining experiments demonstrated that the cytotoxicity induced by 400,600 and 800μg/ml HA -NPs in HepG2 and L02 cells were higher than that in the corresponding control group (P 〈 0.05), and cell cytotoxicity of the same doses of HA - NPs to HepG2 were higher than that of L02 (P 〈 0.05 ). Conclusion High concentrations of HA - NPs have toxic effects both on HepG2 and L02 ceils, but the cytotoxicity of 1_02 cells is lower than that of HepG2 cells at the same concentrations of HA - NPs treatment.
