摘  要:对疑似感染病毒的火龙果(Hylocereus undatus Britt.)进行高通量测序,分析病毒种类和相对含量,为火龙果病毒诊断和检测提供理论基础。测序reads经拼接和注释,共3 969条contig注释为病毒,包含44 354 147条reads。10条contig注释为CVX-NTU(JF937699),其含有的reads占所有病毒reads数量的85.18%;10条contig注释为ZyVX(AY366208),占所有病毒reads数量的4.97%;6条contig注释为SchVX(AY366207),占所有病毒reads数量的3.75%;6条contig注释为CVX(AF308158),占所有病毒reads数量的0.26%。拼接获得的CVX-NTU序列完全覆盖CVX-NTU(JF937699)基因组,核苷酸序列一致性为97%。编码的5个ORF核苷酸序列与CVX-NTU(JF937699)一致性为97%~98%,氨基酸序列一致性为97%~99%。拼接获得的CVX-NTU序列与CVX、ZyVX和SchVX的核苷酸序列一致性分别为78%、75%和70%。这是首次利用高通量测序检测火龙果病毒的报道。A high-throughput sequencing for pitaya (Hylocereus undatus Britt.) tissues infected potentially by virus was performed, and the type and relative content of viruses were studied, which provided a theoretical basis for virus diagnosis and detection of pitaya. On the whole, 3 969 contigs, annotated as virus-related sequence, were produced, from the generated 44 354 147 reads by library sequencing and assembly. Among them, 10, 10, 6 and 6 contigs were annotated as CVX-NTU (JF937699), ZyVX (AY366208), SchVX (AY366207) and CVX (AF308158)-related sequences, respectively. Of all virus-related reads, the count percentage of the CVX-NTU, ZyVX, SchVX and CVX-related reads was 85.18%, 4.97%, 3.75% and 0.26%, respectively. The re-assembled genome sequence of CVX-NTU entirely covered the previously reported CVX-NTU (JF937699)genome. Sequence identity between the two genomes was 97%. The nucleotide sequence identity of 50RFs located in the genome between the re-assembled CVX-NTU and the reported CVX-NTU(JF937699) reached to 97%-98%, and the amino acid identity reached to 97%-99%. The sequence identity of the re-assembled CVX-NTU genome with CVX, ZyVX and SchVX were 78%, 75% and 70%, respectively. This is the first report of virus detection in pitaya using high-throughput sequencing.
