摘 要:目的探讨膜联蛋白A1(Annexin A1)对糖尿病大鼠心功能及肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)的影响。方法将24只SD大鼠随机均分为正常对照组、糖尿病组;利用高糖高脂饮食饲养及较小剂量链脲佐菌素(30 mg/kg)诱导造成2型糖尿病模型,2组分别予以普通喂食、高糖高脂喂食;于第8周末用彩色多普勒超声检测左心室舒张末期容积(LVEDV)、左心室收缩末期容积(LVESV)、二尖瓣舒张早期峰值速度/二尖瓣舒张晚期峰值速度(e/a)比值、左心室射血分数(LVEF)、每搏射血量(SV);第0、4、8周采用实时PCR测定血中Annexin A1表达,采用酶联免疫分析法测定血中TNF-α、IL-1β。结果与正常对照组对比,糖尿病组LVEDV、e/a、SV明显下降,Annexin A1明显上升(P<0.05),8周后糖尿病组Annexin A1显著下降(P<0.01);与正常对照组对比,糖尿病组TNF-α、IL-1β上升明显(P<0.05),8周后上升更明显(P<0.01);糖尿病组Annexin A1与TNF-α、IL-1β负相关(r分别为-0.73、-0.71,P<0.01)。结论 Annexin A1可改善糖尿病大鼠心功能,Annexin A1改善糖尿病大鼠心功能可能与炎症反应有关。Objective To study the effect of annexin A1 on cardiac function, tumor necrosis factor α (TNF-α), and interleukin 1β (IL- 1β) in diabetic rats. Methods Twenty-four SD rats were randomly divided into normal control and diabetic groups. The type 2 diabetes model was induced with a high-glucose and high-fat diet and administration of low-dose streptozotocin.Left ventricular end-diastolic volume (LVEDV), left ventricular end-systolic volume (LVESV), peak velocity of early diastolic mitral-to-late diastolic peak velocity (e/a) ratio, left ventricular ejection fraction (LVEF), and stroke volume (SV) were measured by using color Doppler ultrasonography at the end of week 8. The expression levels of TNF-a and IL- 1β in blood were measured by using enzyme-linked immunosorbent assay, and the expression level of annexin A1 in blood was measured at weeks 0, 4, and 8 by using real-time polymerase chain reaction. Results Compared with the normal control group, the diabetic group had decreased LYEDV, e/a, and SV (P 〈 0.05).The armexin A1 expression level in the diabetic group decreased significantly after 8 weeks (P 〈 0.01 ). The TNF-α and IL-1β levels in the diabetic group were significantly higher than those in the normal control group ( P 〈 0.05 ) and increased significantly after 8 weeks (P 〈 0.01 ). Annexin A1 level correlated with the TNF-α and IL-1β levels in the diabetic group (P 〈 0.01 ). Conclusion Annexin A1 expression shows an anti-inflammatory effect that improved the cardiac function of diabetic rats.
