摘  要:目的探讨miR-92a对人胃癌细胞株生物特性的影响及可能的分子机制。方法在人胃癌细胞(SGC-7901)中瞬时转染miR-92a inhibitor,采用CKK-8法检测细胞株增殖情况,采用TUNEL方法检测细胞凋亡情况,采用Transwell迁移和侵袭实验观察细胞转移情况,并采用Western blot方法检测Bcl-2、Survivin蛋白表达情况。结果人胃癌细胞株在瞬时转染miR-92a inhibitor后,细胞增殖能力明显降低(P<0.05),凋亡明显增加(P<0.05),迁移和侵袭的细胞数量明显较少(P<0.05),Western blot检测结果显示Bcl-2、Survivin蛋白表达水平明显降低(P<0.05)。结论 miR-92a可以促进胃癌细胞增殖、迁移和侵袭,抑制细胞凋亡,其可能通过调控Bcl-2、Survivin来促进胃癌的发生。Objective It is to investigate the effect of miR-92a on the biological characteristics of gastric cancer cell line and its possible molecular mechanism. Methods The miR-92a inhibitor were transiently transfected in SGC-7901, the prolifer- ation circumstance of cell lines was detected by CKK-8 method, the apoptosis circumstance were detected by TUNEL method, the cell migration circumstance was observed by Transwell migration and invasion experiments, the expression of Bcl-2 and Survivin protein were detected by Western blot method. Results After transient transfection of miR-92a inhibitor, the prolifera- tion of SGC-7901 was decreased significantly (P 〈 O. 05), apoptosis was increased significantly ( P 〈 O. 05), the number of the migration and invasion was decreased significantly ( P 〈 0.05 ) , and the results of Western blot showed that the expression of Bcl-2 and Survivin protein were decreased significantly ( all P 〈 0.05). Conclusion miR-92a can promote proliferation, mi- gration and invasion of tumor cells, and inhibit apoptosis, it may promote the development of gastric cancer by controlling Bcl- 2 and Surviving expression.
