摘 要:利用RACE技术从草地早熟禾中克隆赤霉素合成代谢的关键酶GA2-氧化酶基因(PpGA2ox),提交到GenBank,登录号为KX254272。其开放阅读框为1047bp,编码348个氨基酸。系统进化分析表明,PpGA2ox与大麦、小麦及二穗短柄草中的GA2ox亲缘关系较近。亚细胞定位结果显示,其编码的蛋白定位于细胞核和细胞质中。实时荧光定量分析表明,PpGA2ox在幼嫩叶片中的表达量最高,且受GA3、MeJA及IAA三种激素的诱导。To investigate the GA 2-oxidase gene in Kentucky bluegrass, the PpGA 2ox gene was cloned using RACE method in the study. The cDNA sequence was deposited in the NCBI database with accession number KX254272. The open reading frame of PpGA2ox gene was 1047bp, corresponding to 348 amino acids. Phylogenetic analysis exhibited that the PpGA2ox protein was most closely related to those in barley, wheat and Brachypodium distachyon. Subcellular localization results proved that the PpGA2ox pro- tein was localized in both the nucleus and cytoplasm. Real-time quantitative PCR analysis showed that PpGA 2ox gene was expressed most abundantly in young leaves. Moreover, its expression could be induced by exogenous GA3, MeJA or IAA.
