裸燕麦种质遗传完整性SSR分析中样本量的研究

摘　　要：利用SSR分子标记技术分析裸燕麦品种定燕9号和燕科1号的不同样本量群体对遗传参数的影响,确定适宜样本量。以10、20、30、40、50、60、70、80、90为样本量梯度,对不同样本量群体的等位基因数、有效等位基因数、香农指数等遗传参数的变化趋势进行研究,结果表明:用20对SSR引物对定燕9号、燕科1号的90个单株进行扩增,分别检测到43个、32个等位基因,平均每个位点检测到2.09个、1.60个等位基因。样本量大小与等位基因数、多态位点百分率相关。两个材料的样本量分别大于70和50时,等位基因数、多态位点百分率的"S"形拟合曲线变化趋于平缓,其他遗传参数的标准差减小。因此,定燕9号、燕科1号遗传完整性SSR分析的适宜样本量为70和50,确定裸燕麦种质在进行遗传完整性研究时所需的样本量为70。Naked oat Dingyan No.9 and Yanke No.1 of Arena nuda L. were selected as materials mtne study. The genetic parameters of different sample size were investigated by SSR markers. Nine groups of different sample size of naked oat （10,20,30,40,50,60,70,80,90 individuals per group） were set randomly and the number of alleles, the effective number of alleles, Shannon index and other genetic parameters of different sample sizes were studied. The results showed that 20 polymorphic primers were screened from 134 SSR primers, and 43 and 32 alleles were detected in Dingyan No.9 and Yanke No.l, respectively. And number of alleles per locus was 2.09 and 1.60, respectively. The sample size was related to the number of alleles and the percentage of polymorphic loci. In Dingyan No.9 and Yanke No.l, the S curve （the number of alleles and the percentage of polymorphic loci） tended to be gentle when the sample sizes were more than 70 and 50. The standard deviations of other genetic parameters were decreased with the sample size more than 70and 50. Therefore, the suitable sample sizes of the Dingyan No. 9 and Yanke No.1 were 70 and 50 respectively. The appropriate sample size was 70 in the genetic integrity study by SSR for naked oat.
