摘 要:目的制备布洛芬膜控型和骨架型两种缓释微丸,并对体外释放进行分析,研究两者的释药机理。方法两种缓释微丸均采用离心制丸机粉末层积法上药,流化床底喷包衣进行制备,采用Eudragit RLPO作为骨架材料和Eudragit RLPO与E100作为膜控缓释材料。结果微丸光滑圆整,硬度适中,适合包衣,两种微丸均具有明显缓释效果,释药机制为扩散和溶蚀协同作用。结论膜控型缓释微丸释放优于骨架型缓释微丸,制备工艺稳定可行。OBJECTIVE Study on preparation of Ibuprofen sustained-release pellets with membrane and cy- toskeleton controlling mechanism, to analysis the in-vitro drug release behavior of Ibuprofen pellets, and to study the mechanism of drugs. METHODS The sustained-release pellets were prepared by powder lamination process in ten- trifugal pelleting machine and then coated by Fluidized bed. Eudragit RLPO was used as matrix material while Eud- ragit RLPO and El00 as membrane controlled sustained-release material. The release mechanisms of sustained-re- lease pellets were investigated. RESULTS The pellets were smooth and round with moderate hardness, which were suitable for following coating. All pellets had obvious sustained-release effects with the mechanism of diffusion and dissolution. CONCLUSION The release of cytoskeleton-controlling pellets is better than that one. The preparation process of cytoskeleton-controlling pellets was more stable.
