摘 要:目的观察鱼腥草素钠对临床耐药白假丝酵母菌生物膜细胞凋亡的影响。方法利用DAPI染色观察耐药白假丝酵母菌生物膜细胞核的形态;利用FITC-VAD-FMK染色观察耐药白假丝酵母菌生物膜细胞metacaspase活性;分别在JC-1染色和DCFH-DA染色下,采用流式细胞仪检测耐药白假丝酵母菌生物膜细胞内线粒体膜电位(mitochondrial membrane potential,MMP)和活性氧(reactive oxygen species,ROS)水平;利用AnnexinⅤ-FITC/PI染色和流式细胞仪检测耐药白假丝酵母菌生物膜细胞的凋亡率。结果一定浓度的鱼腥草素钠作用下,临床耐药白假丝酵母菌生物膜细胞内ROS水平、生物膜细胞凋亡率、metacaspase酶活性明显升高,MMP水平明显降低,生物膜细胞核出现碎裂和皱缩。结论鱼腥草素钠能够诱导耐药白假丝酵母菌生物膜细胞凋亡。Objective To investigate the effect of sodium houttuyfonate on the apoptosis of Candida albicans biofilm cells with clinical drug-resistance. Methods DAPI staining were performed to observe the morphology of drug-resistant Candida albicans biofilm cell nuclei. FITC-VAD-FMK staining assay was performed to evaluate metacaspase activity in drug-resistant Candida albicans biofilm cells. With the help of JC-1 and DCFH-DA staining, flow cytometry was used to measure mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) level in drug-resistant Candida albicans biofilm cells. Annexin Ⅴ-FITC/PI staining and flow cytometry were used to observe the apoptosis rate of drug-resistant Candida albicans biofilm cells. Results Under the action of sodium houttuyfonate at a certain concentration, drug-resistant Candida albicans biofilm cells had significant increases in ROS level, cell apoptosis rate, and metacaspase activity and a significant reduction in MMP, with fragmentation and shrinkage of cell nucleus. Conclusion Sodium houttuyfonate can induce the apoptosis of drug-resistant Candida albicans biofilm cells.
