百令胶囊影响糖尿病肾病大鼠Caveolin-1蛋白表达机制探讨

摘　　要：目的探讨百令胶囊影响糖尿病肾病大鼠Caveolin-1蛋白表达机制。方法 30只SD大鼠,随机分为对照组(n=10)、模型组(n=20),模型组给予高脂饮食,喂养8周后,模型组大鼠应用链脲佐菌素(STZ)按40 mg/kg腹腔注射,对照组给予等体积枸橼酸钠;72 h测定两组大鼠随机血糖、体质量及24 h尿蛋白定量,将成模的16只大鼠随机平分成2组:糖尿病肾病模型对照组(DN组,n=8)、百令胶囊组(n=8),DN组及正常对照组均给予等量蒸馏水灌胃,百令胶囊组给予百令胶囊(2.5 g/kg·d-1)灌胃,4周后测定各实验组大鼠血糖、24 h尿蛋白定量及Caveolin-1蛋白表达量。结果 DN组Caveolin-1蛋白表达量较对照组升高了(112.8±2.76)%,差异有统计学意义(P<0.05),而百令胶囊组较对照组升高了(61.2±4.31)%,较DN组下降了(51.6±3.48)%,差异均有统计学意义(P<0.05)。结论百令胶囊可能通过调节Caveolin-1蛋白表达发挥对糖尿病肾病肾脏损伤的保护作用。Objective To investigate the effect of Bailing capsule on caveolin -1 protein expression of diabetic nephropathy rats. Methods 30 SD rats were randomly divided into control group （n = 10） and model group （n =20）. The model group was injected Streptozotocin （40 mg/kg） into abdominal cavity after 8 weeks＇high - fat diet and the control group was given same dosage of sodium citrate. After 72 hours, the random blood glucose, weight and 24 - hour urine protein were detected. The 16 model rats were randomly divided into diabetic nephropathy group（DN group, n=8）and Bailing capsule group（n=8）. The DN group and control group were given the same amount of distilled water, and the bailing capsule group was given Bailing Capsule （2.5 g/kg · d-1）. After 4 weeks, the random blood glucose, 24- hour urine protein and Caveolin- 1 protein expression were observed. Results Compared with the control group, the Caveolin-1 protein expression of the DN group was up by （112.8 ±2.76） % and that of the Bailing capsule group was up by （61.2±4.3]） %. Compared with the DN group, the Caveolin - 1 protein expression of the Bailing capsule group decreased by 51.6 ± 3.48 %. The difference was statistically significant （P 〈 0.05）. Conclusion The Bailing capsule may protect the renal injury of diabetic nephropathy by adjusting the Caveolin -1 protein expression.
