摘 要:为了建立高效且遗传性状稳定的大蒜脱毒快繁体系,以宝坻大蒜"六瓣红"的根尖为外植体,探究不同的激素种类及不同浓度配比下,大蒜根尖分生组织区的诱导及不定芽的再分化情况,观察最终诱导出的试管鳞茎的染色体是否异常.研究结果表明,质量浓度为1.0mg/L的2,4-D和1.0mg/L的6-BA共同作用下,根尖诱导形成愈伤组织的诱导率最高,达到了97.5%;质量浓度为1.0mg/L的KT作用下,愈伤组织再分化形成试管苗的再分化率最高,达到了62%.试管苗诱导形成试管鳞茎,诱导率达到了51.3%,检测到试管鳞茎有明显的生长点,且染色体没有异常现象.这些结果均证实本研究成功建立了一种以宝坻大蒜根尖为外植体的高效再生体系.To establish an eliminating virus and rapid propagation system of garlic (Alliium sativum L.) with high efficiency and genetic stability, the root-tips of Baodi "Liu Ban Hong" were used as explants to explore the seduction of root-tips meristematic region and redifferentiation of adventitious bud cultivated in mediums with different kinds of hormones and proportions. The chromosomes of seductive bulblets were examined whether they were abnormal. The results showed that the inducement rate of callus information in root-tips reached the highest value of 97.5% with the co-interaction of 2, 4-D ( 1.0 mg/L) and 6-BA ( 1.0 mg/L). The redifferentiation rate of callus forming tube plantlets reached the highest value (62%) treated with KT (1.0 mg/L). The inducement rate of tube plantlets forming bulblets was 51.3%. The obvious grow- ing points were detected in bulblets of garlic in which there were no chromosome abnormality. All these results demonstrat- ed that a highly-efficient regeneration system of root-tips of Baodi garlic was set up successfully in this research.
