两种植物中喜树碱含量的检测和比较

摘  要:选择乙醇浸提超声波法处理样品,高效液相色谱(HPLC)检测,二极管阵列检测器(DAD),Diamonsil^(TM) C_(18)(4.6×150 mm,5μm)柱,流动相为V_(甲醇):V_水=55:45,流速为0.80 mL·min^(-1),进样量为10μL,柱温为35℃,检测波长为254 nm定量检测.结果表明:马比木中喜树碱的含量高于喜树果,实验结果为药物活性成分提取原料选择提供理论参考.The samples were extracted in ethanol solution by uhrasonic extraction, and the HPLC was used for quantitative determination. The optimum conditions were investigated, in which DiamonsilTM C18(4.6×150 mm,5μm) column was served as separation column, and the mobile phase was methanol: water(55:45) with the flow rate of 0.80 mL·min^-1, while the amount of sample was 10μL, with the column temperature of 35℃ and detection wavelength of 245 nm. Results show that the content of camptothecin in Mappia foetida was higher than that of Common camptotheca fruit. This research will provide good reference for material selection of active ingredient
