摘 要:炭疽芽孢杆菌Bacillus anthracis CGMCC No.12337作为生物催化剂可不对称还原奥卡西平制备抗癫痫药物醋酸艾司利卡西平的关键中间体S-利卡西平.采用响应面优化法对B.anthracis CGMCC No.12337发酵培养基成分和培养条件进行优化,确定最优发酵条件为葡萄糖42g/L,蛋白胨20g/L,初始pH 4.8,磷酸氢二钾0.5g/L,磷酸二氢钾0.5g/L,转速120r/min,温度33℃,接种量10%,培养时间36h.采用该优化培养方法,该炭疽芽孢杆菌产生的羰基还原酶活力达到了775.62U/L,较优化前提高了35.12%.S-licarbazepine was synthesized by asymmetric reduction of oxcarbazepine with Bacillus anthracis CGMCC No. 12337 as catalyst. S-licarbazepine is the key intermediate of antiepileptic drug eslicarbazepine acetate. Response surface analysis method was used to optimize fermentation medium and conditions for B. anthracis CGMCC No. 12337. The optimum fermentation conditions were finally conformed as: 42 g/L glucose, 20 g/L peptone, pH 4. 8, KzHPO4 0. 5 g/L, KH2PO4 0.5 g/L, 120 r/min, 33 ℃, 10% inoculation and 36 h. Under the optimum conditions, the maximum enzyme activity of B. anthracis CGMCC No. 12337 reached 775.62 U/L, with an increase of 35.12% compared to the original fermentation condition.
