摘  要:目的利用DNA条形码技术对泉州口岸常见麻蝇进行鉴定,以期解决麻蝇形态鉴定存在的缺陷问题,以作为形态鉴定的补充。方法通过对泉州口岸常见的5种麻蝇进行PCR扩增、序列测定,进而进行同源性对比、分析进化距离及构建进化树。结果 5种麻蝇细胞色素氧化酶辅酶Ⅰ(cytochrome oxidase subunitⅠgene,COⅠ)基因序列除短角亚麻蝇外,其余种类与Gen Bank中已知序列的同源性均在99%以上;种内差异为0.0%~0.6%(<2%),种间差异则为5.1%~10.0%;进化树显示相同种能聚集在一起,可信度达99%。结论以COⅠ基因作为分子标记可对泉州口岸5种麻蝇进行鉴定,可作为形态鉴定的重要补充。Objective Identification of common Sarcophagidae in Quanzhou port using DNA - barcoding to solve the problem that is limited in identification of Sarcophagidae by morphology. Methods 5 common Sarcophagidae species' CO I gene was amplified and sequenced to alignment, estimate the genetic distances and constructed phylogenetic tree. Results The result of a- lignment with reported sequence in GenBank showed that index of similarity was above 99% except Parasarcophaga brevicor- nis. Intraspecific and interspecific variation ranged from 0.0% to 0.6% ( 〈 2%), and 5.1% to 10.0%, respective- ly. Phylogenetic tree showed that the same species gathered together, bootstrap support was above 99%. Conclusion CO I gene as a molecular marker could be effectively used to identify 5 Sarcophagidae species in Quanzhou port to supplement of morphologi- cally identification.
