苏木乙酸乙酯提取物对心脏移植模型大鼠心肌组织Bcl-2和Bax蛋白表达影响

摘　　要：目的:通过观察苏木乙酸乙酯提取物对同种异位心脏移植急性排斥反应模型大鼠移植凋亡相关蛋白Bcl-2/Bax表达的影响,探讨其免疫抑制作用的机理,为苏木的新药研发提供重要的理论和实验依据。方法:以Wistar大鼠为供体,SD大鼠为受体,制备大鼠同种异位心脏移植模型,造模成功后,采用免疫组织化法检测Bcl-2/Bax蛋白的表达。结果:与模型对照组比较,各治疗组Bcl-2蛋白的表达未见明显改变,差异无统计学意义。苏木组与模型对照组比较,能明显降低Bax蛋白的表达,二者比较有显著性差异,CsA组和苏木加半量CsA组与模型对照组比较,降低Bax蛋白的表达作用更强,有显著性差异;苏木组比CsA组作用弱,二者比较有显著性差异,苏木加半量CsA组比CsA组作用强,但二者比较无显著性差异。结论:在大鼠心脏移植模型中,苏木乙酸乙酯提取物能下调Bax蛋白的表达,从而发挥抑制心肌细胞凋亡的作用。Objective ： Acute heart transplant rejection was investigated with rat model of heterotopic cardiac transplantation. Effect of ethyl acetate extracted from Sappan wood on expression of protein Bcl-2/Bax related with cells apoptosis was observed. The immune suppressive effect mechanism was investigated. These experiments are important theoretical and experimental bases for development of new drug with Sappan wood. Methods ： With an improved Ono method, with Wistar rats as donors and SD rats as recipients, model of heterotopic cardiac transplantation of rats was established. Afler the model was made, expression of Bcl-2/Bax was detected with lmmunohistochemistry （ IHC ） . Results ： After compared with the model control group, there was no significant change of the Bel-2 protein expression of treatment groups. There was no statistical significant difference between them. After compared with the model control group, there was significant decrease of Baxprotein expression of the CsA group, with significant difference between them. After compared with the model control group, there was more significant decrease of Bax protein expression of the CsA group and Sappan wood added with half CsA group, with significant difference between them.Effect of the group of Sappan wood on reducing Bax protein expression was weaker than the CsA group, and there was significant difference between them. Effect of the group of Sappan wood added with half CsA on reducing 13ax protein expression was better than the CsA group, however there was no significant difference between them. Conclusion ： Ethyl acetate extracted from Sappan wood can cut the expression of Bax protein and play the role of inhibiting myocardial apoptosis in the heterotopic cardiac transplantation.
