针刺对哮喘大鼠肺组织中Eotaxin、RANTES表达的影响

摘　　要：目的:探讨针刺对支气管哮喘大鼠肺组织中Eotaxin、RANTES表达的调控。方法:将24只SPF级SD大鼠随机分为空白组、哮喘模型组、哮喘模型针刺组,每组8只。各组分别给予相应干预后,采用HE染色法观察大鼠肺组织中病理变化,免疫荧光染色法检测肺组织中Eotaxin、RANTES的表达,ELISA检测大鼠BALF及血清中Eotaxin、RANTES表达的程度。结果:哮喘模型针刺组大鼠肺组织病理改变较哮喘模型组明显减轻,与空白组差别不明显;哮喘模型组Eotaxin、RANTES蛋白表达高于哮喘模型针刺组(P<0.05);哮喘针刺组BALF及血清中Eotaxin、RANTES含量均低于哮喘模型组(P<0.05),但与空白组差异不明显(P>0.05)。结论:哮喘大鼠BALF及血清中Eotaxin、RANTES的表达水平明显增高;针刺治疗可以明显降低其表达水平,提示针刺可以通过调节Eotaxin、RANTES的表达来减轻气道炎症反应。Objective： To examine the mechanism of acupuncture on c-fos expression in the lung tissues of asthmatic rats. Methods： Twenty-four SPF male SD rats were randomly divided into 3 groups with 8 rats in each group. Groups were given accordingly interventions. Light microscope was used to observe the effects of acupuncture on Eotaxin,RANTES expression with Cell counting,HE staining and Immunofluorescence method. Enzyme-linked immunosorbent assay was used to measure the levels of Eotaxin and RANTES in bronchoalveolar lavage fluid and serum. Results： HE staining of rat lung indicated that the pathological changes of acupuncture group were slighter than the asthmatic model group＇s,and similar to the blank group＇s. Immunofluorescence showed that the protein expression of Eotaxin and RANTES in lung tissue in acupuncture group were lower than that in the asthmatic model group（ P〈0. 05）. Enzyme-linked immunosorbent assay showed that the protein expressions of Eotaxin and RANTES in bronchoalveolar lavage fluid and serum in acupuncture group were lower than that in the asthmatic model group（ P〈0. 05）,but similar to the blank group（ P〈0. 05）. Conclusion： In asthmatic rats BALF and serum Eotaxin and RANTES expression levels were significantly higher. Acupuncture can decrease the expressions of Eotaxin and RANTES,suggesting that acupuncture can reduce airway inflammation by regulating the expressions of Eotaxin and RANTES.
