摘 要:目的:通过观察肾衰饮对慢性肾功能衰竭大鼠肾脏bmp-7、pdgf、timp-1 mRNA表达的影响,探讨肾衰饮延缓慢性肾功能衰竭的药效学机制。方法:52只SD大鼠随机分为正常组、模型组、尿毒清组及肾衰饮组。采用腺嘌呤灌胃法制作大鼠肾衰模型。造模成功后肾衰饮组大鼠给予肾衰饮煎剂灌胃,尿毒清组大鼠给予尿毒清颗粒灌胃,各组大鼠连续给药4周,全自动生化分析仪检测血肌酐(SCr)、尿素氮(BUN)及白蛋白水平,HE染色及Masson染色观察各组大鼠肾组织病理变化,Real time-PCR技术检测大鼠肾脏bmp-7、pdgf、timp-1 mRNA表达情况。结果:与正常组比较,模型组血肌酐、尿素氮水平显著升高(P<0.01),白蛋白水平显著降低(P<0.01);与模型组比较,肾衰饮组和尿毒清组大鼠血肌酐、尿素氮水平均显著降低(P<0.01);白蛋白水平均显著升高(P<0.01)。HE及Masson染色显示模型组大鼠肾脏组织发生明显的病理学改变,肾衰饮组及尿毒清组大鼠肾脏病理改变发生明显改善。与正常组相比,模型组大鼠肾脏bmp-7基因mRNA表达水平显著下降(P<0.01),pdgf、timp-1基因mRNA表达水平显著升高(P<0.01);与模型组比较,肾衰饮组大鼠肾脏bmp-7基因mRNA表达水平显著升高(P<0.01),pdgf、timp-1基因mRNA表达水平显著下降(P<0.01)。结论:肾衰饮可能通过调节慢性肾功能衰竭大鼠肾脏bmp-7、pdgf、timp-1 mRNA表达,发挥其延缓慢性肾功能衰竭进展的重要生物学作用。Objective: According to observing the effect of Shenshuaiyin on the expressions of bmp-7,pdgf and timp-1 in kidney tissues of chronic renal failure rats,to discuss the pharmacology mechanism of the Shenshuaiyin delaying the renal interstitial fibrosis. Methods: Fifty-two SD rats were randomly divided into control group,model group,Shenshuaiyin group and Niaoduqing group. Use adenine gavage method for renal failure model in rats. After the success of the building,the Shenshuaiyin group rats were given Shenshuaiyin by gavage. The Niaoduqing group rats were given Niaoduqing by gavage. Every groups' rats were given drugs for 4 weeks continuously. Use the automatic biochemical analyzer to test the SCr,BUN and albumin levels. Observe the changes of the kidney morphology by HE and Masson dyeing. Real time-PCR tested the expressions of bmp-7,pdgf and timp-1 mRNA of the rats' kidney. Results: Comparing with the control group,the levels of serum creatinine and urea nitrogen increased significantly( P〈0. 01) and the level of albumin decreased significantly( P〈0. 01) in model group. Comparing with the model group,the levels of serum creatinine and urea nitrogen decreased significantly( P〈0. 01) and the level of albumin increased significantly( P〈0. 01) in Shenshuaiyin group and Niaoduqing group. HE and Masson dyeing showed that there were obvious pathological changes in the kidney tissue in model group. The pathological changes of kidney were improved obviously in Shenshuaiyin group and Niaoduqing group. Comparing with the control group,the expression levels of bmp-7 mRNA decreased significantly( P〈0. 01) and the expression levels of pdgf and timp-1 mRNA increased significantly( P〈0. 01) in model group. Comparing with the model group,the expression level of bmp-7 mRNA increased significantly( P〈0. 01) and the expression levels of pdgf and timp-1 mRNA decreased significantly( P〈0. 01) in Shenshuaiyin group.
