摘 要:目的:通过对黄芪水煎液多成分肠吸收暴露特征的分析,探讨黄芪多成分在大鼠体外肠吸收以及体内代谢情况。方法:采用翻转肠囊法和腹主动脉取血等试验,结合HPLC指纹图谱,研究黄芪水煎液化学成分在肠吸收与体内代谢情况。结果:通过翻转肠囊实验,明确了16种通过肠吸收的成分,其中成分1、2、4、6(毛蕊异黄酮葡萄糖苷)、7(芒柄花苷)、11以原型成分被肠吸收。成分M1、M2、M3、M4、M5、M6、M7、M8、M9、M10是通过肠壁代谢的新成分。HPLC分析腹主动脉采血明确了8种成分入血,其中M9为可通过肠壁代谢吸收入血的成分,成分R1、R2、R3、R4、R5、R6、R7为经过代谢吸收入血的新成分。结论:综合肠吸收、入血成分分析,肠道和肝脏对黄芪水煎液化学成分具有一定代谢作用,十二指肠段与回肠段是对黄芪成分吸收较好的部位,空肠段是代谢的主要部位,且肠道和肝脏存在明显的吸收代谢差异。Objective: To study the intestinal absorption and metabolism of Radix Astragalus Decoction in rats in vivo and in vitro. Methods: To investigate metabolism of the chemical constituents of Radix Astragalus Decoction. The gut sacs was carried out as well as the blood was taken out from abdominal aorta and all samples were analyzed by HPLC. Results:There were 16 constituents absorbed by intestine of rats and 8 constituents were detected in the blood. Compound 1,2,4,6( calycosin-7-O-β-D-glucoside),7( Ononin) and 11 were absorbed in prototype forms in the intestine directly.Compound M1,M2,M3,M4,M5,M6,M7,M8,M9 and M10 were new ones. On the other hand,8 compositions[M9,R1,R2,R3,R4,R5,R6,R7]could be absorbed into blood through analysis serum samples obtaining from aorta abdominalis of rats. The M9 could be absorbed directly by intestine,while R1,R2,R3,R4,R5,R6 and R7 were absorbed into blood in new form. Conclusion: Based on the intestinal absorption experiment and analysis of compsition in blood,intestine and liver have a certain effect on the chemical composition of Astragalus decoction. Duodenal and ileum segments are the better parts of Astragali Radix absorption and the jejunum segment is the main part of metabolism. The absorption and metabolism differences between intestine and liver were significant.
