摘 要:目的:通过观察黄芪多糖对肠缺血再灌注(IR)损伤幼鼠肠组织肿瘤坏死因子α(TNF-α)、细胞间黏附分子1(ICAM-1)、白介素-6(IL-6)表达以及T淋巴细胞亚群分布的影响,探讨黄芪多糖对幼鼠肠缺血再灌注损伤肠组织的保护作用。方法:选用4周龄SD大鼠60只,随机分成假手术组、缺血再灌注组(IR组)及黄芪多糖干预组(APS组),各20只。假手术组仅暴露腹腔脏器,不做任何干预;缺血再灌注组及黄芪多糖干预组,阻断肠系膜上动脉造成缺血60 min,缺血再灌注组注射0.5 m L生理盐水,黄芪多糖干预组注射20 mg/100 g黄芪多糖。分别于再灌注后30、60、90、120 min每组处死5只大鼠取小肠组织进行免疫组织化学染色,检测小肠中TNF-α、ICAM-1及IL-6的表达,流式细胞仪检测小肠CD_3^+、CD_4^+、CD_8^+T淋巴细胞百分比,计算CD_4^+/CD_8^+比值。结果:正常肠组织中的TNF-α、ICAM-1及IL-6呈现出低水平表达,缺血再灌注模型、黄芪多糖干预组肠组织中的TNF-α、ICAM-1及IL-6含量与假手术组比较均显著升高(P<0.01),缺血再灌注模型组及黄芪多糖干预组肠组织中TNF-α、ICAM-1及IL-6含量均从30 min开始逐渐升高,至90 min时达到顶峰,之后逐渐下降。黄芪多糖干预组肠组织中TNF-α、ICAM-1以及IL-6含量在各个时间点与缺血再灌注模型组比较,均显著降低(P<0.01)。与假手术组和黄芪多糖干预组比较缺血再灌注组各个时间点小肠CD_3^+、CD_4^+T淋巴细胞百分比均显著下降(P<0.05),缺血再灌注组CD+8T淋巴细胞百分比与假手术组和黄芪多糖干预组比较各个时间点均显著升高(P<0.05),CD_4^+/CD_8^+比值,缺血再灌注组与假手术组和黄芪多糖干预组比较均显著下降(P<0.01)。结论:黄芪多糖能够有效减轻肠组织缺血再灌注损伤,可能与下调TNF-α、ICAM-1及IL-6的表达及调节T淋巴细胞亚群的平衡有关。Objective: To disscuss the protective effect of astragalus polysaccharides on intestinal tissue ischemia-reperfusion injury on young rats by observing the effect of astragalus polysaccharides on expressions of TNF-α,ICAM-1 and IL-6 in intestinal tissue of the young rats. Method: Sixty healthy male SD rats with 4 weeks old were randomly divided into three groups: sham-operated group,ischemic reperfusion model group and astragalus polysaccharides group,each group 20 rats. The sham-operated group was only exposure to intra-abdominal organs,without any other intervention. The schemic reperfusion model group and astragalus polysaccharides group blocked arteries on mesenteric for 60 min. The ischemic reperfusion model group injected 0. 5 m L physiologic saline and the astragalus p olysaccharides group injected 20 mg/100 g astragalus polysaccharides. All animals were sacrificed by 30,60,90 and 120 min. And then small intestine was taken out to test the TNF-α,ICAM-1 and IL-6 concentrations. The proportions of CD3^+,CD4^+,CD8^+T lymphocytes in intestine were determined with flow cytometer and the proportions of CD4^+/CD8^+ was calculated. Results:The sham-operated group's rats had low TNF-α,ICAM-1 and IL-6 expressions. The TNF-α,ICAM-1 and IL-6 expressions of sham-operated group were significantly lower than the other two groups'( P〈0. 01). The immunohistochemical staining showed that the TNF-α,ICAM-1 and IL-6 contents in intestinal tissue from the beginning of reperfusion 30 min gradually increased to reache a peak in 90 min in three groups and then declined. At each time point in hemia-reperfusion group and astragalus polysaccharides group,TNF-α,ICAM-1 and IL-6 concentrations in intestinal tissue were significantly higher than those of the sham-operated group( P〈0. 01). At each time point,the proportions of CD3^+ and CD4^+ T lymphocytes in intestine on sham-operated group and astragalus polysaccharides group were significantly lower than that of the ischemia-reperfusion group�
