针刺对去势雌兔干眼症模型的实验研究

摘　　要：目的:观察针刺眼周五穴对去势雌兔干眼症模型的炎症因子及黏蛋白MUC5AC、MUC19的影响。方法:60只新西兰雌兔,随机分为空白对照组(A),针刺对照组(B),针刺组(C)和模型对照组(D),每组15只。针刺组和模型对照组采用双侧卵巢切除术制造去势雌兔干眼症模型,其中针刺对照组和针刺组给予针刺睛明、攒竹、阳白、丝竹空、瞳子髎,模型对照组给予针刺眼周空白穴。针刺每周3次,4周为1个疗程。分别于治疗前、治疗后1周、2周及4周收集泪液检测IL-6、TNF-α、LF、MUC5AC、MUC19的浓度,并于治疗后采用荧光免疫组织化学法检测结膜组织中MUC5AC、MUC19的表达。结果:针刺后,针刺组较治疗前IL-6、TNF-α明显降低,LF、MUC5AC、MUC19明显升高,差异具有统计学意义(P<0.05,P<0.01),与模型对照组比较,差异具有统计学意义(P<0.05,P<0.01)。治疗后模型对照组结膜MUC5AC、MUC19总阳性率显著低于空白对照组,差异具有统计学意义(P<0.01),针刺组结膜MUC5AC、MUC19总阳性率较空白对照组总阳性率比较,差异无统计学意义(P>0.05),但明显优于模型对照组(P<0.01)。结论:针刺治疗可以抑制IL-6、TNF-α的促炎活性,增加LF水平达到抑菌作用,从而抑制眼表的炎症反应;另一方面通过促进杯状细胞及睑板腺对黏蛋白的合成和分泌,提高泪液中黏蛋白的表达,恢复泪膜的稳定性,从而恢复眼表的正常结构和功能。Objective： To observe the effect of inflammatory factors on ovariectomized female acupoint needle dazzling Friday rabit dry eye model and influence the expression of MUC5 AC and MUC19; Methods： 60 New Zealand female rabbits were randomly divided into control group（ A）,acupuncture group（ B）,acupuncture group（ C） and model group（ D） 15 rats in each group. The acupuncture group and model control group with bilateral ophorectomy made in ovariectomized rabbit dry eye model,the acupuncture group and acupuncture group were given acupuncture Jingming,zanzhu,Yang Bai,sizhukong,Tong Zi Liao,the model control group was given acupuncture eye week point blank. Acupuncture3 times a week,4 weeks for a course of treatment. Respectively before and after treatment in 1 weeks,2 weeks and 4weeks to collect tear detection of IL-6,TNF-alpha,LF,MUC5 AC,MUC19 concentration,expression of conjunctival tissue were measured by immunohistochemistry assay after treatment in MUC5 AC and MUC19; Results： after acupuncture,acupuncture group compared with before treatment,IL-6,TNF-alpha was significantly reduced,LF,MUC5 AC,MUC19 increased significantly,the difference was statistically significant（ P〈0. 05,P〈0. 01）,compared with the model group,the difference was statistically significant（ P〈0. 05,P〈0. 01）. After treatment,the model control group,conjunctival MUC5 AC total positive rate of MUC19 was significantly lower than the control group,the difference was statistically significant（ P〈0. 01）,acupuncture group,conjunctival MUC5 AC total positive rate of MUC19 compared with the blank control group the positive rate,the difference was not statistically significant（ P〉0. 05）,but was significantly better than the model control group（ P〈0. 01）; Conclusion： acupuncture treatment can inhibit IL-6 and TNF-alpha proinflammatory activity,increase LF level to achieve antibacterial effect,thereby inhibiting the inflammation of ocular surface; on the other hand,by promoting the secretion and synthesis
